Papaverine is one of the drugs that can enhance tissue expansion. But it has not been widely used in clinic because of lack of effective and easy way for delivery. Based on the review of related studies, we propose that tissue expansion may be enhanced by papaverine through external using.To evaluate the permeability of papaverine hydrochloride cream, a method was established for determination of papaverine hydrochloride in blood and skin with spectrophotometer. Papaverine cream was externally used on skin tissue of pig and human in vitro and in vivo, and the concentration of papaverine in the tissue was detected.In this study, papaverine was extracted using chloroform-hexane as the solvent from blood and skin tissue. In vitro, the papaverine concentration was 90.5 — 259.6μg/g in skin tissue of pig and the absorption rate was 1.72~6.52% within 12 hours when 0.017 ~ 0.045g/cm~2 papaverine cream was externally used. The papaverine concentration in human skin tissue was 23~143.9μg/g and the absorption rate was 2.5~4.7% within 12 hours when 0.012~0.047g/cm~2 papaverine cream was used on human skin. In vivo, the papaverine concentration was 33.32~97.08μg/g in pig skin within 1—8 hours when 0.4g/cm~2 papaverine cream was externally used.Mini pigs were used to evaluate the effect of external using of papaverine on tissue expansion rate, blood flow and survival of expanded flaps.
