Background: Diabetic nephropathy(DN)is one of the most serious complications of diabetes(DM).With the development of society and economy,more and more people suffered from DM,consequently,the morbidity of DN increased, therefore, the number of patients with chronic renal failure due to DN also increased.So,the study about mechanism of DN is becoming very importment.Proteinuria is the mark of DN and its forms is due to glomerular filtration barrier injury.Recent studies showed that podocyte and some molecules(nephrin, podocin) made pivotal contribution to maintenance the selective of filtration barrier of the normal glomerulus, podocyte injury is associated with marked proteinuria.ACEI and ARB can reduce proteinuria of DN by preventing damage of podocyte. Rosiglitazone,a novel insulin-sensitizing agent,have been reported that it can ameliorate proteinuria of DN,but the exact mechanism remains unclear.Objective:The main aim of this study was to investigate the changes about podocytes ultrastructure, proteinuria and the expression of nephrin and podocin in DN rats,and the relationship among these changes, observe the effects of rosiglitazone and losartan influence on podocyte.Methods:35 Wistar rats were divided at random into normal control group(N,n=10) , DN model group(D,n=10) , treatment group with rosiglitazone(R,5mg/ Kg .d,n=10) and losartan(L,50mg/ Kg .d,n=10) and...
