Objective To investigate comprehensively differential gene expressive profile among different labor situation in human myometrium in order to scan out labor-associated genes . Differentially expressive genes were tested both gene and protein level in myometrium and peripheral blood from pregnancy women and studying their significances.Methods cDNA microarry technology : The RNA was extracted from myometrium in corpus and lower position of thirty pregnant women , and synthesized into cDNA, then labeled with cy3/cy5 respectively.The labeled cDNA was hybridization with human cDNA microarray of 8064 genes, and the differential expression spots of cDNA were identified by Array Vision and MIDAS software. Scanning of differential expression genes by searching in the Human date base was alone via http: //www.ncbi.nlm.nih.gov and http: //www.chgc.sh.cn, then differential expression genes were picked out and established differential gene expressive profile in between corpus and lower position form nature active labor,between corpus and corpus from nature active labor and no-labor,between corpus and corpus from oxytocin induced labor and no-labor.RT-PCR : The differential expression genes, high expressive heat shock 70kD protein 1B( HSPA1B) and lower expressive voltage-dependent Calcium channel-Lα1c (CACNA1C) were verified using semi-quantitative reverse transcription polymerase chain reaction in the...
