Objective:1 , To investigate the inhibitory effects of Qidan granule on collagen accumulation and discuss the mechanism of Qidan granule in treatment of pulmonary fibrosis, by observed the changes of tissues hydroxyproline and effects of VG dyeing in pulmonary tissues of bleomycin-induced pulmonary fibrosis rats treaded by Qidan granule or hybridization.2,To observe the changes of angiogenesis in bleomycin-induced pulmonary fibrosis and the connection of angiogenesis and pulmonary fibrosis. Angiogenesis was evaluated by microvessel density , vascular endothdlial growth factor and hypoxia-inducible factor-1α in lung tissues by immunohistochemistry and electron microscopy. To explore the relation from VEGF to MVD and HIF-1α in pulmonary fibrosis . Methods:1,160 pathogen-free Sprague-Dawley (SD) rats were randomly divided into six groups: normal control group(N), model group (M), Qidan granule group 1 and 2(Q1, Q2) , Hydrocortison group1 and 2(H1, H2). Normal group was treated with saline water. Pulmonary fibrosis model was induced by intratracheal instillation of bleomycin (5mg/kg body weight).40 rats used as model group and other rats were treated with Qidan granules (3125mg/kg/d) or treated with Hydrocortison (25mg/kg/d), respectively. Rats were sacrificed at 7,14,28 or 42 day post-bleomycin.Then the lungs were harvested and fixed in paraformaldehyde and embedded in paraffin wax.
