The acupoints have been widely used in clinical practice ever since they were recorded. More and more doctors of the following generations have used these acupoints to cure diseases based on the clinical needs. This dissertation makes a comprehensive and systematic research on their medical effects on insomnia, and then, takes a further study on the effect of Sishencong curing insomnia mice and the adjusting and controlling mechanism of monoamine neurotransmitter in brain-stem reticular formation of insomnia rats at the molecular level in the light of the results of the study of the past medical literature and clinic application related to Sishencong (EX-HN1), taking insomnia as the definite research direction.Objective: Investigate the sleeping effects of Sishencong (EX-HN1) and the adjusting and controlling mechanism of monoamine neurotransmitter in brain-stem reticular formation for insomnia.Methods: Conclude and sum up the clinic application condition by the literature research on Sishencong; Detect the synergism about the sleeping effects of Sishencong and Nembutal sodium in mice by using routine pharmacological methods; Duplicate insomnia model of rats by means of PCPA abdominal injection, Study the results of acupuncturing Sishencong by observing the content of NE in insomnia rats' LC, the content of 5-HT in insomnia rats'NRD, the expression of 5-HT1A receptor and 5-HT2a receptor in insomnia rats' DRN.Results: 1. Sishencong can cure many kinds of diseases, the retrieval rate of insomnia is the first one in the diseases cured by Sishencong. 2. Acupuncturing the acupoints, combined with Nembutal sodium, are able to accelerate sleep, shorten the latent period of sleep; prolong the sleep duration; increase the sleep number of the mice, compared with the normal group the difference is notable (P<0.05). 3. Abdominal injection of PCPA makes the normal rats into the insomnia condition, the content of 5-HT in NRD decreases, the content of NE in pontine LC increases, Acupuncturing Sishencong can decrease the content of NE in pontine LC, increase the content of 5-HT in NRD, compared with the control group the difference is notable (P<0.05;P<0.01). 4. After abdominal injection of PCPA, the expression of 5-HT1A receptor in NRD of the rats weakens, the expression of 5-HT2a receptor strengthens, compared with the normal group the difference is notable (P<0.05); Acupuncturing Sishencong can enhance the expression of 5-HT1A receptor of the insomnia rats' NRD (P<0.01), the electro-acupuncture group is better than the hand-acupuncture group (P<0.05), weaken the expression of 5-HT2a, compared with the control group the difference is notable (P<0.05;P<0.01), the electro-acupuncture group is better than the hand-acupuncture group(P<0.05).Conclusion: 1.the retrieval research indicates that the scope of the clinic application of Sishencong should be comprehensive, and Sishencong should be effective acupoints to cure insomnia, could be able to cure many kinds of diseases, among these dieases, Sishencong curing insomnia is the first one and Sishencong is the main acupoint for curing insomnia. 2. Acupuncturing Sishencong can be able to accelerate sleep, shorten the latent period of sleep; prolong the sleep duration; increase the sleep number of the mice. 3. Acupuncturing Sishencong can decrease the content of NE which increased in pontine LC; increase the content of 5-HT which decreased in NRD, 4. Acupuncturing Sishencong can enhance the expression of 5-HT1A receptor of the insomnia rats' NRD; weaken the expression of 5-HT2A receptor of the insomnia rats' NRD.
