Carpesium triste Maxim is the species of Carpesium L. genus of the family Compositae, widely distributed in Heilongjiang, Jilin, Liaoning, Sichuan and Yunnan provinces and Xizang autonomous region, etc. The whole plant has been used as herbal medicine for the treatment of furuncle, mastitis, sore throat, toothache, urinary tract infection and diarrhea. Two new sesquiterpene lactones from C. triste var. manshuricum were reported by Korean chemists.Ligularia narynensis and Ligularia dolichobotrys Diels are perennial herbaceous plants of Ligularia Cass. (Compositae). L. narynensis mainly distributed in Tianshan Mountains of Xinjiang Uygur Autonomous Region and former Soviet Union. Latest chemical investigation of this species found two peptides by Chinese chemists. L. dolichobotrys Diels mainly distributed in the north and south of Qinling Mountains. Our group investigated chemical composition in it, and found one eremophilenolide and one stigmasterol, recently.Dracocephalum heterophyllum Benth is the species of Dracocephalum L. genus of the family Labiatae, widely distributed in Shanxi, Gansu, Sichuan and Tsinghai provinces, Inner Mongolia, Ningxia hui autonomous region, Xizang autonomous region, Xinjiang Uygur Autonomous Region, etc. The aerial part of the species used to treat hypertension, lymphadenitis, cough caused by dryness-heat.In the present paper, we studied the chemical constituents of C. triste, L. narynensis, L. dolichobotrys and D. heterophyllum. The chemical compounds were isolated by means of silica gel column chromatography (CC), preparative thin layer chromatography (PTLC) and recrystallization etc. The structures of these compounds were elucidated by chemical transformations and spectroscopic methods including IR, UV, EI-MS, FAB-MS, HR-ESI-MS, 1H-NMR, NOEDS, 13C-NMR, DEPT, 1H-1H COSY, 1H-1H NOESY, HSQC, HMBC, CD etc. From C. triste, twenty-seven compounds were isolated, fourteen of them are new compounds. From L. narynensis, twenty-seven compounds were isolated, including nine new compounds. Fifteen and twenty-three compounds were isolated from L. dolichobotrys and D. heterophyllum, respectively.Furthermore, the in vitro cytotoxic activities of some compounds against human hepatoma cells (SMMC-7721), human promyelocytic leukemia cells (HL-60) and human hepatocytes cells (L02) were carried out by sulforhodamine B (SRB) method. The 50 % inhibitory concentration value (IC50) showed that compound 1.1-1.7 had significant cytotoxic activity against the HL-60 cells, that compound 1.12 had significant cytotoxic activity against SMMC-7721 and HL-60 cells, and that compound 2.2 and 2.4 had cytotoxic activity against the HL-60 cells.Results of this investigation were expected to afford important academic basis for the structure-activity relations, Pharmacology, screening new medicine, chemotaxonomy and utilization of natural resource.Finally, oplopane-type sesquiterpenes naturally found in 1966-2006 were reviewed in the present thesis.
