Chinese medicine has a good effect in reducing chemotherapy side effects.This article in two parts on the YQFYT combined with chemotherapy in alleviating side effects and increasing Synergistic effect of chemotherapy,and inhibition of cell proliferation, and Promoting the role and mechanism of apoptosis, etc, associated clinical and experimental research will be conducted.Object:1 To Observation Synergism and Attenuation of chemotherapy of clinical of YQFYT on colorectal cancer patients After surgery.2 To Study the effect of YQFYT containing serum on colorectal Lovo Proliferation and Apoptosis, and to explore its molecular mechanism of apoptosis.and whether YQFYT combined with chemotherapy drugs have a better effect.Methods:1 Clinical Observation:Since June 2009 to January 2010,selected hospital chemotherapy after radical resection of colorectal cancer patients with 53 cases,after excluding 4 cases,evaluation of patients with 49 cases,24 cases in treatment group, control group 25 cases;YQFYT for the treatment group to take, and body weight, toxicity, immune function and KPS score Will be observed,after chemotherapy.2 Experimental Study:the Restrain effect of Lovo colorectal cancer cell of YQFYT, chemotherapy drugs,YQFYT+ chemotherapy drugs each containing serum, serum control group and blank control group will be test by MTT assay;flow cytometry to detect cell cycle and apoptosis;immunoblotting (Western blotting) to detect cell-related apoptosis protein. Results:1 Clinical Observation:After chemotherapy, Treatment group was better than the control groupj in Stability of body weight and KPS score, with significant difference, p<0.05);the treatment group so significantly better than the control group,in Leukemia reduced,Platelet reduced and nausea and vomiting, diarrhea and ALT increased of Toxicity,P<0.01; However, no significant difference between the two groups in Increased level of bilirubin, ALP, AST, Cr, BUN and neurotoxicity,(P>0.05);After chemotherapy, CD4+, CD8+,CD4+/CD8+ and NK of two groups was decreased, but CD4+ and CD4+/CD8+of the treatment group was significantly higher, with statistical significance, P<0.01.2 Experimental Study:MTT assay showed that the Restrain effect of Lovo colorectal cancer cell of YQFYT compared with the control group was significantly different, P<0.01,showing tumor suppressor role in aging, dose enhancement; YQFYT containing serum group increased the proportion of Gl phase cells,S phase cells decreased compared with the control group were statistically significant, P<0.01;)Apoptosis rate in colorectal cancer Of YQFYT containing serum group was significantly higher,with significant difference, P<0.01;bcl-2 IOD value Of YQFYT containing Serum group was decreased, but bax IOD value rise control group statistically difference,P<0.01;Inhibition of colon cancer cell proliferation Of YQFYT combined with chemotherapy drugs group is better than two drugs alone, and have significant differences, P<0.01,Cell cycle regulation and apoptosis, bcl-2 and bax IOD value effects have a similar effect.Conclusion:1 Synergism and Attenuation of YQFYT Was found on Colorectal cancer patients receiving postoperative chemotherapy.2 YQFYT containing serum have proliferation inhibition on Lovo Colorectal cancer,and combined with chemotherapy drugs is better.3 YQFYT containing serum have proliferation inhibition on Lovo Colorectal cancer through adjusting its cell cycle and promote apoptosis achieved.4 YQFYT containing serum has Induction of apoptosis on lovo colorectal cancer mainly through up protein expression Of bax, down protein expression Of bcl-2 to achieve.
