Purpose:.To clarify the inhibitory effect of DCC-2036 on the proliferation,invasion and migration of colon cancer LoVo cells.Experimental design: In the first part,in vivo experiments verified the effect of DCC-2036 on the proliferation,apoptosis,invasion and migration of colon cancer LoVo cells.Colon cancer cell line LoVo was used as the research model in the experiment.Flow cytometry was used to detect the change of apoptosis of LoVo cells after DCC-2036 treatment.Transwell,clone formation,and scratch tests were used to detect the proliferation,Changes in invasion and migration capabilities;Western Blotting detected the change of EMT markers in LoVo cells after DCC-2036 treatment;The second part,the experiments in vivo verified that the effect of DCC-2036 on the growth of LoVo cell xenografts in nude mice.In the experiment,nude mice transplanted with LoVo cells were used as a research model.The growth of tumors after DCC-2036 treatment was detected by measuring and statistically analyzing the changes in tumor size;HE staining and immunohistochemistry were used to detect DCC-2036 treatment.Differences in morphology and proliferation index of transplanted tumor under microscope Result: 1.It has been proved by MTS experiment that DCC-2036 can inhibit the cell viability of LoVo cells.2.Annexin V-FITC / PI dual staining flow cytometry showed that DCC-2036 can induce apoptosis of LoVo cells.3.Through clone formation experiments,it was proved that DCC-2036 can significantly inhibit the proliferation of LoVo cells.4.Transwell experiment verified that DCC-2036 can inhibit the invasion of LoVo cells.4.Through scratch test,it was proved that DCC-2036 can significantly inhibit the migration of LoVo cells.5.In vivo experiments in mice demonstrated that DCC-2036 can inhibit the growth of LoVo cell-derived xenografts in mice.Conclusion: DCC-2036 induces the apoptosis of colorectal cancer LoVo cells,and inhibits its proliferation,invasion and migration.
