Objective:To research the effect of electro acupuncture Baihui through Qingding in the treatment of PD and the mechanism of that on dopaminergic neurons in the repair protection.Methods: In this study C57BL / 6 strains of mice 100, 10 to 11 weeks of age, body weight 20±2g, were randomly divided into control group, the model group, EA group and Madopar group, n = 25. Model group, EA group and Madopar group were given intraperitoneal injection of MPTP (0.03mg / g body weight), once every day for 7 days Preparation of mouse model of PD; control group received normal saline intraperitoneally. Of the 8th day, EA group selected Baihui through Qianding the EA frequency 2 ~ 100HZ, voltage 1V, on time, every 20min, for 14 days; Madopar group were given Madopar suspension gavage (0.12 mg / g body weight), once every day for 14 days; control group and model group were given saline intraperitoneally. Mice in each group before the climbing pole, hanging behavioral observation; experimental mice were killed on day 22, take the substantia nigra detect brain GSH content and GSH-PX activity reflects the loss of dopaminergic neuronal degeneration, and to the light and Under the morphology observed by immunohistochemical method nigra Bcl-2 and Bax expression to reflect apoptosis; observation and in situ hybridization BDNFmRNA expression and integrated optical density, using reverse transcription polymerase chain reaction Nestin measured change in the number of detected NSC proliferation and differentiation. SPSS13.0 software applications available to the data analysis and processing.Results: 1. Hanging experiments and climbing pole test score: model group score lower EA group and Madopar group mice score higher. 2.GSH and GSH-PX activity detection: model group in the substantia nigra of mice were significantly decreased GSH, GSH-PX activity decreased; GSH of EA group and Madopar group were significantly increased and GSH-PX of them activity increased. 3.Apoptosis observations: Bcl-2 immunoreactive cells observed: with the control group; EA and Madopar group increased; Bax immunoreactive cells observed: the model group increased significantly; EA and Madopar group was significantly reduced. 4. BDNF mRNA expression observed: the number of positive cells and the integral optical density results: the model group have reduced; the EA group and Madopar group have increased; The EA group and Madopar group of BDNF mRNA positive cells showed no differences; integrated optical density is a significant difference comparison. 5. Nestin mRNA expression observed: average optical density results: EA group and the normal group, model group and Madopar group, Nestin mRNA expression were significantly increased. Madopar compared with model group, there was no significant difference.Conclusion: EA Baihui through Qianding at the PD model mice can improve the behavior disorder; to substantia nigra neurons increased antioxidant capacity, suppression of free radicals; EA can inhibit the dopaminergic neurons apoptosis; induced increase BDNFmRNA expression of PD model mouse; promote NSC activation, proliferation, differentiation, and promote nerve cell compensatory restoration, suggesting that EA Baihui through Qianding can be achieved on the PD mice substantia nigra dopaminergic neurons in the repair and protective effect, with better treatment.
