PART ONEOBJECTIVE : this study was designed to investigate the continuous changes of the concentrations of ET-1 , TNF , E-SLT and IL-8 in plasm of rat lung model of edema ,and to observe the therapeutic effects of lung edema with miltiorrhizae and the possible action mechanism .METHODS : the rat lung model of edema was prepared by being exposed to chlorine (3000ppm) for 10 minutes . the rats was randomized into control (n=20) and lung exposed to chlorine groups,the latter group was further randomized into untreated lung edema group (n=20), multiorrhizae group (n=20). Blood specimens and lung tissue sample were collected in different testing periods. Wet and dry weight of right lung were calculated . double antibody sandwich quantitative ELISA method was used to measure the concentration of E-SLT , and radioimmunoassay (RIA) were used to measure the concentrations of TNF , ET-1 and IL-8 . RESULTS : compared with those in normal control group , the plasma concentrations of E-SLT , ET-1 , TNF and IL-8 in animals exposured to chlorine were increased significantly in multiorrhizae group , the plasmconcentrations of E-SLT , TNF , ET-1 and IL-8 were lower than those in untreated lung edema group . The relationship analysis showed that the ratio of lung water had significantly positive relation with those cytokines. CONCLUSIONS : E-SLT , TNF , IL-8 can cause Lung edema , multiorrhizae can reduce lung water by decreasing the plasma concentrations of E-SLT , ET-1 , TNF and IL-8 .PART TWOOBJECTIVE :To study the effects of ET-1 , TNF , E-SLT and IL-8 in plasm of 24 patients with lung contusion .METHODS : double antibody sandwich quantitative ELISA method was used to measure the concentration of E-SLT , and radioimmunoassay (RIA) were used to measure the concentrations of TNF , ET-1 and IL-8 in 24 patients with lung contusion and 10 normal control subjects .RESULTS : compared with those in normal control group ,the plasma concentrations of E-SLT ,ET-1 ,TNF and IL-8 in lung contusion were increased significantly . there were also significant differences of the concentration of ET-1 ,TNF ,E-SLT and IL-8 in 24 patients between pretreatment and posttreatment .CONCLUSIONS : ET-1 , TNF , E-SLT and IL-8 might be involved inlung contusion by causing lung edema . Assay of ET-1 , TNF ,E-SLT and IL-8 is very useful on observing advance of lung contusion and judging therapeutic .
