Objective:1、To discuss the ERK pathway and its relative genetic function in animal and cell models of Lewis lung carcinoma through Ginsenoside Rg3treatment.2、To discuss the prevention ability of Ginsenoside Rg3in A/J mice lung tumors animal model through Ginsenoside Rg3treatment.Methods:1、The Lewis lung carcinoma mice animal model was established. After feeding Rg3to the mice by gavage for3weeks, the volumes of tumors were calculated.2、The inhibition effect on Lewis lung carcinoma cell line was detected through MTT assay. The changes in Dusp6, Bax, Bcl-2expressions in tumor model were detected through Realtime-PCR. The changes in ERK and p-ERK protein expression were determined by Western blot.3、Benzo(a)pyrene-induced Lung Tumorigenesis in A/J mice animal model was established. After feeding Rg3to the mice by gavage for18weeks, the volumes and multiplicity of the tumors were calculated.Results:1、Ginsenoside Rg3inhibits Lewis lung carcinoma in both cell and animal models (P<0.01).2、ERK protein expression was significantly decreased in the Rg3group compared to the control group as determined by Western blot assay. Dusp6and Bax were overexpressed in Rg3group compared to the control (P<0.01), while the Bcl-2was underexpressed as determined by Realtime-PCR assay (P<0.01)3、Ginsenoside Rg3both inhibits and prevents lung carcinoma growth in the A/J mice animal model.Conclusion:1、Ginsenoside Rg3exhibits certain inhibition ability in both in vivo and in vitro models of Lewis lung carcinoma.2、Ginsenoside Rg3can significantly inhibit pulmonary adenoma formation and growth in A/J mice, showing certain inhibition and prevention functions in the in vivo model of A/J mice lung tumors.
