268 Prevention And Control Of Anti-liver Fibrosis In Experimental Rat Liver Fibrosis

Objective.To study the curative and preventive effect of 286 on chronic hepatitis ,and to probe the mechanism of the effect primarily. Method: Different concentration of 286 in high dose and loe dose was used to the rat models which made by damaging the liver with the simple CL4.At the same time another group of rats was for the comparison are treated with colchicine.The degree of liver fibrosis was dicided by the tissue through Masson dye with optic microscope.The ALT.AST and r-GT was chosen to evaluate the degree of liver damage.The ALB.GLOB.TP and A/G was done toestimate the degree of liver store function.The weight of liver ,heart,spleen,lung,kidney and spermary.the index of liver ,the degree of liveliness,the self-defence consciousness,the dining and the fur tenderness of rat.the mortality of rats received CL4 and so on.which were assessed to else change in rats du ring the experimentation.FN and LN(the non-collagenous index) out of ECM were measured by immuno-histochemistry mean, and the accumulation of ECM was measured by type I amd III coHagen(the collagenous index), a -SMA was chosen as a symbol ef .the activated reposit fat cell.TGF- P1 was chosen as a symbol of th ieccellular factor made the reposit fat cell activated.We observed the mi sclnanism of 286 through the variety of the upper indexes during the experiment. Result:286 restrain the secretion of TGF- P 1 and decrea se the quantity of a -SMA in liver cells ,and the accumulation of, HSJ.LN, type I and HI collagen in ECM The degree of liver cell damai ie was alleviated and the adverse effect what the purtenance received was lightened The store function of liver was enhanced The preventive and curative effect on chronic hepatitis was re ached. Between thequantity-effect relation,the high dose was better than the low dose.Between the time-effect relation.the preventive drug served was the better.Conclusion:286 can prevent and treat the chronic hepatitis effectively.The mechanism is related to repressing secretion of TGF-P1,reducing the reposit fat cell activation and decreasing the accumulation; of ECM.
