Objective: Traditional Chinese Medicinal theory suggested that Benign Prostatic Hyperplasia (BPH) has correlation with Chronic Prostatitis (CP) in clinical. Qianlie Huichun can treat CP,we think that it also can treat BPH. To deteck the content of Testosterone (T) , Estradiol (E2 ), Dihydrotestosterone (DHT),the expression of Fas, Proliferating cell nuclear antigen (PCNA), Vascular endothelial growth factor (VEGF),cell apoptosis rate ,and observe the microstructure and ultrastructure change in rats' prostate constitution. Studying the Pathogenesis of BPH in cytokine, protein,cell form structure levels partly, to reveal the therapeutical effect of Qianlie Huichun.Methods: S. D male rats were divided into 6 groups : normal group, model group, Estradiol group, Qianlie Huichun low dose group, middle dose group and high dose group. Rats were castrated ,then subcutaneous injected testosterone propionate for 30 days and treated with each group's drug at the same time. After 30 days , the rats was put to death, separate the prostate constitution to observe and deteck: (1). Observe the changes of prostate weight , prostate exponent, histology under light microscope of each group. (2). Observe the changes of ultrastructure under electon microscope. (3). Take the radioimmunochemistry to measure the sex hormone content of prostate constitution. (4) .Use the method of immunohistochemistry to detect the expression of Fas PCNA, VEGF in each prostate constitution. (5). Take the Flow Cytometry to measure the apoptosis rate of each group.Results: (1).The protate weight, epithelia cell height increased significantly (P<0. 01) ; (2).Model rats have obvious anomaly in histology and ultrastructure in prostate, Qianlie Huichun can relieve the anomaly. (3).The contents of dihydrotestosterone (DHT) in model rats increased greatly ( P<0. 01 ) , Qianlie Huichun can low the level of DHT. (4).The expression of Fas in model prostate cells decreased significantly, but the PCNAVEGF increased greatly, Qianlie Huichun can adjust the expressions. (5).The apoptosis rate of prostate cell in model rats decreased significantly (P<0. 01 ) , Qianlie Huichun can increased the apoptosis rate of prostate cell.Conclusions: The study has suggested that BPH has relation with T, DHT' increasement , E2' decreasement , the hign expression of PCNA, VEGF and low expression of Fas ,cell apoptosis reduced in prostate constitution. Qianlie Huichun can decrease the content of T, DHT and the expression of PCNA, VEGF, increased the expression of Fas and the apoptosis rate of prostate cell in prostate constitution .The research suggested that Qianlie Huichun also can treat BPH,meanwhile reveal the he therapeutical effect of Qianlie Huichun in whole,cell and molecullevels.
