1. ObjectiveIdiopathic thrombocytopenic purpura(ITP) is a kind of hemorrhage disease .Its episode cause isn't clear. The aim of study is to explore the formula-syndrome relationship that ZIDIANKELI(ZDKL) and Idiopathic thrombocytopenic purpura (Qi deficiency and blood stasis syndrome).First, review the study of ITP syndrome and pathological research of Qi deficiency and blood stasis syndrome; Second, discuss prescribing rules of ZIDIANKELI We suggest the pathological basis of ITP (Qi deficiency and blood stasis syndrome)and a threatment target of ZDKL by experiment study of pharmacodynamics ,in vivo and medicated serum on ZDKL.2. MethodsTo build the mouse model of chronic ITP, we applied the method of immunity, that is APS was injected into BALB/c mouse by intraperitoneal injection. Divided the group by the platelet count randomly. To explore the change rules of the formation of the animal model by the method of immunity with the general accepted standard that Qi deficiency and blood stasis. We treated the syndrome with ZIDIANKELI and used the result to disproof the change of Qi deficiency and blood stasis in the animal model; Discussing the relationship and mechanism between ZIDIANKELI and chronic ITP the prescription and syndrome from the four different aspects of the whole body organs cell molecule and by using two methods of in vivo and in virvo.3. Results3. 1. Pharmacodynamical indexes of ITP3. 1. 1.The effects on peripheral platelet of ITP model mouseThe peripheral platelet number of model group was reduced (P < 0.05 ) .The clotting time was longer (P < 0.05 ) .The quantities of CD41, CD61 were reduced (P < 0.05 ) . The number of bone marrow megakaryocyte and immature megakaryocyte was increased,or platlet-producing megakaryocyte was decreased (P < 0.05 ) .There were significant differences of them in between control group and model group. The bleeding of ITP is related to the number, the aggregating function and the structure of the platelet. At the same time ,the mature of bone marrow megakaryocyte is blockedThe large-dose group of ZDKL and prednisone acetate group are superior to other groups in producing the platelet (P < 0.05 ); The large-dose group of ZDKL decreases the clotting time and increases the quantity of the platelet membrane glycoproteins; The large-dose and middle- dose groups of ZDKL inhibit the producing of immature megakaryocyte and develop the producing of platelet-producing megakaryocyte. The large-dose group of ZDKL is superior to other groups. 3. 1. 2. Pharmacodynamical indexes of Qi deficiency blood stasis syndrome Model groups were injected with APS. After 48h ecchymosis appears in the abdominal and tails.on 5th day, model mice appear activity and weight decreasing (P < 0.05 X thin hair, short time of swimming (P < 0.05 ); ZDKL can alleviate all of them; the large-dose group of ZDKL is the best 3. 2. The effects on microstructure of glomerulus's vascular endothelial cells of UP modelmouseThe changes of vascular endothelial cells in model groups are outstanding. The irregular change out of nuclear, and the distribution of heterochromatin with the ball shape in the nuclear. The mitochondrion can be seen in the cytoplasm, the dilataltion of the HER and the exfoliation of the surface ribosome. All the proofs manifested the changes of the endothelium in cell morphology in ITP. To compare with the blank group, the group of ZDKL has no obvious change. It indicates the ZDKL can correct the changes of the endothelium in cell morphology in ITP.3. 3. The effects on vascular endothelial factors of ITP model mouseThe quantity of peripheral blood endothelin (ET-1) was increased (P < 0.05). The quantity of calcitonin gene-related peptide (CGRP ) was't changed (P > 0.05 ) .There were significant differences of them in between control group and model group. ET-1/ CGRP was increased.
