Bone allograft is widely used in orthopaedics and related disciplines, although its osteoinductive capacity is much less satisfactory compared with the autograft. Moreover, the allograft material should be subjected to sterilization with the danger of transmitting infection inherent in it, which in turn leads to further diminution in osteoinductive power. How to improve the osteoinductivity remains a problem hard to resolve in the research of bone allografting. Proceeding from the experiences of the authors who had previously worked on processing of allograft material, we developed a reconstituted bone allograft. The product was prepared by combining the human cancellous bone granules which had undergone defatting, partial decalcification and deproteinization, with human bone morphog-enetic protein(hBMP) which possesses high osteoinductivity. The recombinant which was sterilized with ethylene oxide has been used in repairing bone defects in animal experiments and in clinical practice with encouraging preliminary results. The reconstituted bone allograft was found to possess strong osteoinductive power without appreciable immunogenicity and the harzard of transmitting diseases, which made it a new grafting material more suitable for use as a substitute for autograft. The authors have found a better solution to the major problem of bone allografting having weak osteoinductive power and the harzard of transmitting human diseases.BMP and the theory of bone induction is the most active and rapidly progressing aspect in the study of bone grafting with their results and conclusions becoming the theoretical basis for guiding the clinical practice in bone graf(?)ing.
