Yeast extract paste(40g·kg-1 ig) or potassium oxonate (300mg·kg-1 ip) was used respectively to establish mice hyperuricemia model.Sodium urate injection was given to form rats acute gouty arthritis model.TF composition was used intragastrically in both the mice hyperuricemia model and the rats acute gouty arthritis model in different dosis (mice:high dose-7.00g·kg-1·d-1, middle dose-3.50g·kg-1·d-1, low dose-1.75g·kg-1·d-1; rats:high dose-3.50g·kg-1·d-1, middle dose-1.75g·kg-1·d-1, low dose-0.88g·kg-1·d-1 ),and then the level of uric acid inserum and the other index that related with pain and inflammation were detected,while compared with Benzbromarone,Allopurinol and Indometacin. Results: TF composition was able to elicit dose-dependent hypouricemic effects and the xanthine oxidase level was reduced. It also shown the improvement of the tumefaction on the toe of rats and the ear of mice, and the decrease of the pain excitatory transmitter (K+, DA, 5-HT) and the proinflammatory cytokines (TL-1β,IL-6,IL-8,TNF-α,PGE2) in the rats peripheral cavitas articularis.Pathological examination shown the inflammatory cell infiltration and the monosodium urate crystals in the joint which induce acute attacks of gouty arthritis inflammation was inhibited. Electron microscope shown the inflammatory reaction was decreased and the cellular membrane and cellular organ were stabilized. Conclusion: TF composition was shown to possess potent hypouricemic effects,to inhibit the monosodium urate crystals deposited in the joint,to protect the ultramicro structure in synovium of joint,and antiinflammation and analgesia through improve uric acid ejection,inhibit xanthine oxidase activity and the pain excitatory transmitter and the proinflammatory cytokines releasing.
