Objective:1. To establish the rat model of atherosclerosis with the compound method.2. Observation the influence of kangmai capsule on the the atherosclerosis model in rats through the level of the blood fat (includ total cholesterol、triglyceride、high-density lipoprotein cholesterol), low density lipoprotein cholesterol (LDL-C); Tumor necrosis factor (TNF alpha) and interleukin 10 (IL-10), oxidized low density lipoprotein (ox-LDL), VCAM 1), and the expression of NF-kB in diseased tissue.Methods:60 healthy SD rats were divided into normal control group 10 (give only normal diet) and model group 50, the Model group were given a high-fat diet+intraperitoneal injection of vitamin D3+propylthiouracil (lavage)+immune damage+FeSO4 (water) to produce atherosclerosis modell month after 50 rats were randomly assigned into the pathologic model group (no treatment), kangmai capsule high, medium and low dose treatment group and plug pulse treatment group, each group have10 rats;Every Group continue to give a high-fat diet and start to give drugs:kangmai capsule high-dose group:3.12/Kg, medium-dose group 1.56/Kg, low-dose group 0.78/Kg, Tong plug pulse piece group:1.56/Kg, diluted with normal saline into 10 ml/Kg, and were filled to the stomach, once per day. Model group: normal saline 10 ml/Kg to fill the stomach, once per day. After 3 weeks, all rats were put to death. Light and electron microscope each rat aortic morphology change, fully automatic biochemical instrument direct detection of blood lipid levels, using ELISA method to detect each group rats before and after drug serum TNF alpha, interleukin 10 (IL-10), ox-LDL, the concentration of the adhesion factor (VCAM 1); Use of immunohistochemistry and Western blot method to detect the expression of NF-kB P65 diseased tissue.Results:1.This animal model were confirmed successfully by pathological detection.2. Kangmai capsule treatment group can significantly reduce the degree of Arteriosclerosis, decrease the thickness of aorta vascular intima, increase the middle membrane of Vascular. Compared with the pathologic model group, the high and medium dose of kangmai capsule group (P< 0.01), low dose group (P< 0.05);the high dose group compared with tong plug pulse tablet group (P< 0.01).3. The serum CHO, LDL and HDL levels increased significantly of pathologic model group, kangmai capsule and tong plug can obviously decrease it, compared with the pathologic model group (P< 0.01).4. Compared with the pathologic model group, kangmai capsule group can significantly reduce serum TNF alpha, ox-LDL, VCAM 1, elevated serum levels of IL-10 (P< 0.01); Kangmai capsule high-dose group compared with tong plug group is significant difference (P< 0.01).5. Compared with the pathologic model group, kangmai capsule and tong plug group have decreased the level of NF-kB P65 obviously (P< 0.01); Kang capsule high dose compared with tong plug group significant difference (P< 0.01).Conclusion:1.The improved method can successfully establishe the atherosclerosis rat model, which can be used to judge the relationship between inflammatory markers and atherosclerosis.2.Kangmai capsule inhibit the development of atherosclerosis by lowering inflammatory factor, for example TNF-a、ox-LDL, VCAM 1, and increase anti-inflammatory factor IL-10.3.The NF-kB of Atherosclerotic plaques rised, and involved in the formation of atherosclerosis, kangmai capsule can reduce the expression of NF-kB protein in diseased tissue, thus effectively inhibit atherosclerosis progression.
