| Objective:This paper is for the pharmacodynamics of external use Xiang Lian Jin Huang San for treating Yang-syndrome ulcers by both internal and external experiment as well as clinical research. Besides we do the exploration for sake of giving some evidences for more efficient uses of this powder in clinical application, which will also give some thinking for the deeper research on TCM external therapy and medicine.Method:①Using AGAR plate method to observe vitro antimicrobial activity of the single herbs and all parties of Xiang Lian Jin Huang San;Using the guinea pigs skin infections caused by staphylococcus aureus as the animal models of Yang-syndrome ulcers to observe the conditions for ulcers bump regression, the descending of the infl-ammatory skin tempreture and the total number of white blood cells in peripheral blood after treating with Xiang Lian Jin Huang San.②Using the clindamycin phosph-ate gel as the contrast to observe the therapentic effects of Xiang Lian Jin Huang San for the acne vulgaris with the lesions like red, swollen, hot and pain.③Using the imm-unohistochemical method to determine the effect that Xiang Lian Jin Huang San made on the content of IL-1, IL-6, IL-8,TNF-a, IFN-y protein of the Yang-syndrome ulcers of model animals’inflammatory skins; Prepare the transdermal liquid of Xiang Lian Jin Huang San, regarding the berberine hydrochloride as the standard;Compare the Xiang Lian Jin Huang San transdermal liquid with its powder for the vitro antimicro-bial activity and the effect for treating Yang-syndrome ulcers of guinea pigs skin infections caused by S.aureus.Result:①The single herbs and all parties of Xiang Lian Jin Huang San have the better vitro antibacterial activity to the tested Gram-positive bacteria than those Gram-negative one;Houpu has the best bacteriostatic effect of staphylococcus aureus,while Muxiang and Gancao take the second place; Chenpi has a perfectly bacteriostatic eff-ect of Escherichia coli,pseudomonas genus,salmonella,and klebsiella,followed by Bai- zhi.Compared with model control group,even Xiang Lian Jin Huang San high and medium dosage groups of guinea pig skin swelling subsided significantly(P<0,01); Xiang Lian Jin Huang San high and medium dosage groups of guinea pig skin temperature decreased significantly(P<0.01); Xiang Lian Jin Huang San’s total dose groups of guinea pigs in the peripheral blood leukocyte decreased obviously(P<0.01); Compared with Ru Yi Jin Huang San group,high dose group of guinea pig skin bump subsidise more obvious(P<0.05).ⅢCompared with untreated,the integral decreased in both Xiang Lian Jin Huang group and clindamycin gel group after treating the acne vulgaris with the lesion like red,hot,swllowen and painful,and the differences are both have significant change(P<0.01);After treatment,the Symptoms sores between the two groups have no significant difference(P>0.05);there is no significant difference betw-een those two groups of therapeutic effect(P>0.05).③Compared with model control group,the IL-1protein in the guinea pig skin tissues in high dose of Xiang Lian Jin Huang San group expresses significantly decreased(P<0.01);the IL-6protein in the guinea pig skin tissues in medium dose of Xiang Lian Jin Huang San group expresses significantly decreased(P<0.01);the IL-8protein in the guinea pig skin tissues in high, medium dose of Xiang Lian Jin Huang San group expresses decreased(P<0.05);the TNF-a protein in the guinea pig skin tissues in high,medium dose of Xiang Lian Jin Huang San group expresses decreased(P<0.05);the IFN-y protein in the guinea pig skin tissues in high.medium dose of Xiang Lian Jin Huang San group expresses decreased,but there is no statistically significant difference(P>0.05).By the skin in vitro of test animal,we can produce Xiang Lian Jin Huang San transdermal liquid,and it contains the same Rf value fluorescent yellow spots compared with berberine hydrochloride by Camera bell ows uv transmission apparatus test;Both Xiang Lian Jin Huang San and Xiang Lian Jin Huang San transdermal liquid have the similar functions of bacteriostatic action in vitro activity to8types26strains of the common pathogenic bacteria and quality control bacteria in clinic; Xiang Lian Jin Huang San transdermal liquid has the function of diminishing swelling and lowering the temperature of the inflammatory skin of Yang-syndrome as well as decrease the total WBC in the peripheral blood cells. Conclusion:①The single herbs and all parties of Xiang Lian Jin Huang San have the bacteriostatic action in vitro activity; Xiang Lian Jin Huang San has the function to relieve swelling and decrease the inflammatory skin temperature; Xiang Lian Jin Huang San has the effect that can reduce the total number of WBC in peripheral blood of infected guinea pigs.②Xiang Lian Jin Huang San has a good clinical therapeutic effect for the acne vulgaris with red,swollen,hot and pain lesions.③Xiang Lian Jin Huang San maybe play the role for clearing heat and relieving toxin,removing the swollen by descending the expressions of inflammatory cytokines(IL-1,IL-6,IL-8,TN F-a);Xiang Lian Jin Huang San transdermal liquid has the similar or even better therapeutic effects than Xiang Lian Jin Huang San itself.Xiang Lian Jin Huang San plays its role when treating externally by passing through the skin. |
