Trichosanthes kirilowii Maxim., a kind of perennial herbaceous climbing plant,belongs to the gourd family. Its fruits, peels, seeds, and roots which were known as Gualou,Gualoupi, Gualouzi and Tianhuafen in Chinese have been widely used in traditionalChinese medicine (TCM) because of the important phamaceutical values. Trichosantheskirilowii Maxim. was first recorded in Shen Nong’s Herbal Classic. Gualoupi is sweet inflavor and cold in nature. Gualoupi attributes to lung meridian and stomach meridian. Ithas been widely used in TCM for many years to treat many diseases such as cough withlung heat, phelegm turbidity, chest stuffiness and pain, and thoracic fullness due to theabilities to clear heat and dissipate phlegm, relieve stuffiness of the chest and dissipatemass. Previous pharmacological studies revealed that it has strong activities such asdilating the coronary artery, eliminating phlegm, antibacterial, and anticancer etc. Gualoupiparenteral solution was made using Gualoupi as raw materials, and it was used to treatcoronary heart disease and stable angina pectoris due to its abilities of promoting qi andrelieving stuffiness, opening chest and dissipating mass. In spite of many studies have beenreported recently, the active components have not been revealed and the quality controlsystem needs to be improved. In order to further study the chemical components of thepeels of Trichosanthes kirilowii Maxim., the chemical composition was systematicallystudied in the thesis, which can supply some reference for clinical application, qualityspecification formulation and development of the natural resource.1. Separation, purification and identification of nucleosides in the peels ofTrichosanthes kirilowii Maxim.In order to investigate the water-soluble constituents of the peels of Trichosantheskirilowii Maxim., the separation and purification were performed by variouschromatography methods including macroporous absorption resin column chromatography and semi preparative high performance liquid chromatography. Nine known compoundswere isolated and the structures were determined by spectroscopic analysis. They wereidentified as cytosine (â… ), uracil (â…¡), hypoxanthine (â…¢), guanine (â…£), xanthine (â…¤),adenine (â…¥), guanosine (â…¦),6-isoinosine (â…§) and adenosine (â…¨), among whichcompound â… , â…¢, â…£, and â…¤ were isolated from this plant for the first time.2. Separation, purification and identification of flavonoids in the peels ofTrichosanthes kirilowii Maxim.In order to investigate the constituents of ethyl acetate fraction of the peels ofTrichosanthes kirilowii Maxim., the separation and purification were performed by variouschromatography methods including polyamide resin column chromatography and semipreparative high performance liquid chromatography. Seven known flavonoids wereisolated and the structures were determined by spectroscopic analysis. They were identifiedas quercetin-3-O-[α-L-rhamnose (1â†'2)-β-D-glucopyranosyl]-5-O-β-D-glucopy ranoside(â… ), quercetin-3-O-rutinoside (â…¡), apigenin-7-O-β-D-glucopyranoside (â…¢),diosmetin-7-O-β-D-glucopyranoside (â…£), luteolin (â…¤), apigenin (â…¥), and diosmetin (â…¦),among which compound â… , â…¡, â…¢, â…¤ and â…¥ were isolated from this plant for thefirst time.3. Separation, purification and identification of fatty acids and sterols in thepeels of Trichosanthes kirilowii Maxim.In order to investigate the constituents of light petroleum fraction of the peels ofTrichosanthes kirilowii Maxim., the separation and purification were performed byrepeated silica gel column chromatography and recrystallization. Six known compoundsincluding three fatty acids and three sterols were isolated and the structures weredetermined by spectroscopic analysis. They were identified as methyl stearate (â… ),palmitic acid methyl ester (â…¡), palmitic acid (â…¢), α-spinasterol (â…£), stigmasterol (â…¤),and β-sitosterol (â…¥).4. Separation and identification of volatile oil in the peels of Trichosantheskirilowii Maxim.The essential oil from the peels of Trichosanthes kirilowii Maxim. was extracted bySoxhlet method and its chemical constituents were analyzed by gas chromatograph-massspectrometry (GC-MS).60compounds in the essential oil were isolated and identified andthe relative contents of the components in the essential oil were determined by peak-area normalization. The compounds identified accounts93%of the essential oil. The majorcomponents are fatty acids (29.73%), aldehydes (26.81%), alkynes (14.38%), alkenes(9.11%), esters (4.64%), alcohols (2.13%), ketones (2.53%) and other aromatic compounds(3.68%).5. Separation, purification and identification of alkaloids in the peels ofTrichosanthes kirilowii Maxim.In order to investigate the alkaloids constituents of the peels of Trichosantheskirilowii Maxim., acid extraction and alkali precipitation method was used to extractalkaloids and the separation and purification were performed by various chromatographymethods including reversed phase C18column chromatography and preparative highperformance liquid chromatography. Five compounds were isolated and two of which wereidentified as adenine (â… ) and6-isoinosine (â…¡). And the other three compounds have notbeen identified.
