Purpose:In this experiment, the cell model, the expression of aquaporin AQP-4 and occludin protein in cirrhosis hepatic encephalopathy brain edema observed in brain tissue by immunohistochemistry and western blot method, combined with hepatic encephalopathy praxiology liver function (ALT, AST, TBIL), blood ammonia (BA), the nerve cells oncosis, changes in cerebral tissue, and to investigate the effect of eliminating toxic Du xiao Gan qing wan in rats on cerebral edema hepatic encephalopathy thus further confirmed Du xiao Gan qing wan treatment of hepatic clearance and its mechanisms of hepatic encephalopathyMethod:1.The rats were randomly divided into six groups:control group, model group.the lactulose group,the lowm,edium,high dose of Duxiao Ganqing wan.2.Using Zhao qing Zhu and other reference solvent carbon tetrachloride to give 50% each per 0.5ml·100g-1,2 times·wk-1, continuous subcutaneous 12wk and joint (high-fat low-protein foods with 10% ethanol as the only beverage) the modeling method for preparing a composite factor cirrhosis model, combined with the use of in the first 13wk by 250 mg/kg TAA intraperitoneally every other day for 2 consecutive injections of the modeling process for the preparation of cirrhosis experimental animal models of hepatic encephalopathy3.After the modeling,in addition to the model group and the normal group.the other groups were given the Duxiao Ganqing wan and lactulose therapy.4.Research on the impact of school administration through the water maze test and grading and other observations Duxiao Ganqing wan about liver cirrhosis in rats with hepatic encephalopathy behavior in 7 days.5.Removing Duxiao Ganqing wan impact on the pathological tissue and liver cirrhosis rat brain model of hepatic encephalopathy by electron microscopy poison.6.By immunohistochemistry and western blot experiments to observe Du xiaoGan qing wan to eliminate the impact of liver cirrhosis of hepatic encephalopathy model expression in rat brain tissue of occludin protein.7.By immunohistochemistry and western blot experiments observe Duxiao Ganqing wan to eliminate eliminate the impact of liver cirrhosis of hepatic encephalopathy model expression in rat brain tissue of AQP-4 protein.Result:1. Description eliminate Duxiao Ganqing wan cirrhosis hepatic encephalopathy due to learning and memory impairment does have some improvement.2)Consumers Du xiao Gan qing wan in rats with hepatic encephalopathy model of neural reflexes and grading:Staging reflex hepatic encephalopathy in rats by the low of Duxiao Ganqing wan treatment can be significantly improved and reduced, especially the high and middle dose group of Du xiao Gan qingwan, reduce the role of hepatic encephalopathy staging and model significant difference between groups.2.Consumers Du xiao Gan qing wan on hepatic encephalopathy model of rat brain:Lactulose group,Du xiao Gan qing pillof high-dose,medium-dose and low-dose group, hepatic encephalopathy, cerebral edema reduced, especially when more obvious in high doses,suggesting that drug elimination from the pathological Duxiao Ganqing wan on animal models of hepatic encephalopathy and induced cerebral edema by the protective effect.3.Consumers Du xiao Gan qing wan hepatic encephalopathy rat liver function,blood ammonia effects:Compared with the model group,the treatment group were significantly decreased serum indexes (P<0.05),the high dose of Du xiao Gan qing wan in liver function (ALT, AST, TBIL), decreased blood ammonia,a significant difference (P<0.01),explanation of Du xiao Gan qing wan on HE rat liver function (ALT, AST, TBIL), blood ammonia significant improvement.4.Consumers Duxiao Gan qing wancirrhosis hepatic encephalopathy rats with nerve cells Oncosis effects:Compared with the model group,Duxiao Ganqing wan of the high,medium and low dose group, western medicine group performance on the cell morphology of the cell body shrinkage,nuclear condensation, chromatin condensation,etc.oncosis performance,while stained brown or brown particles,their number,morphology were lower than the model group.5.Eliminate toxic effects on Duxiao Ganqingwan expression of occludin in liver cirrhosis hepatic encephalopathy rat brain model:Immunohistochemistry and Western Blot results further proved cirrhosis hepatic encephalopathy occludin protein expression is closely related to permeability changes and distribution of vascular endothelial cells,increased vascular permeability brain when it lowered.when Hepatic encephalopathy happen cerebral vascular permeability further increase,suggesting that low expression of occludin may be tight junctions (tight junctions,TJ) the molecular basis of structural damage.From the molecular level to further prove Duxiao Ganqing pill cleared by clearing the liver eliminate ammonia and other toxins,reduce blood-brain barrier damage,increased expression of occludin protein, cerebral edema,so as to achieve the purpose of the treatment of hepatic encephalopathy6.Eliminate toxic effects on Du xiao Gan qing wanexpression of AQP-4 protein in liver cirrhosis hepatic encephalopathy brain tissue model:AQP-4 protein immunohistochemistry and Western Blot results are consistent indirectly proved by the experiment,the degree of hepatic encephalopathy in patients with cirrhosis of cerebral edema is closely related to the expression of AQP-4 protein that appears to provide a theoretical cirrhosis cerebral edema basis,thus providing a new theoretical basis and clinical pathways for the treatment of Duxiao Ganqing wan eliminate liver cirrhosis hepatic encephalopathy treatment of cerebral edema.Conclusion:l.Using solvent carbon tetrachloride to give 50% each per 0.5ml·100g-1,2times·wk-1, continuous subcutaneous 12wk,combined with250mg/kgTAA intraperitoneally every other day for 2 consecutive injections of modeling methods liver cirrhosis hepatic encephalopathy animal model successfully prepared.2.The results of this experiment show that the observed indicators:Du xiao Gan qing wan can improve liver clearance behavior changes of rats with hepatic encephalopathy,reduce the level of BA and restore liver function, affects the brain pathology morphological changes of nerve cells inhibit Oncosis inhibiting AQP-4 protein expression and protein occludin,cerebral edema,so as to achieve the clinical treatment of hepatic encephalopathy role.3.Duxiao Ganqing wan can be cleared by buqijianpi and Ziyinqingre,diarrhea drug dredge in order to achieve reduction in the absorption of toxins,protect the liver,brain,intestinal therapeutic purposes,and is worthy of further research and development.4.Through theoretical and experimental studies,we further proof of" brain-gut Tongzhi" theory,combined with Tong fu xie re logic,application Duxiao Ganqing wan on liver cirrhosis hepatic encephalopathy treatment is safe and effective, worthy of promotion.
