Objective:1?To investigate the expression and clinical significance of PGAM1 in salivary adenoid cystic carcinoma;2?To study the effect of PGAM1 gene on biological characteristics of salivary adenoid cystic carcinoma;To investigate the mechanism of PGAM1 in the development of salivary adenoid cystic carcinoma.Methods:1?PGAM1 expression was detected by immunohistochemistry in 31 cases of SACC and 25 normal glands,and the correlation with the clinical pathology parameter was analyzed;2?The highly metastatic human salivary adenoid cystic carcinoma cell line(SACC-LM)was transient transfected by PGAM1 si RNA interference clips.The interference efficiency was examined by q RT-PCR and Western-blot;Cell cycle changes was determined with flow cytometry;Cell proliferation was evaluated by CCK8;Cell migration was examined by wound-healing assay;cells invasion was observed by transwell;the changes of MMP-2,MMP-9,N-cadherin,E-cadherin and vimentin expression before and after the interference by Western-blot.Results:1?PGAM1 was expressed at a low level in 25 normal glands.In 31 cases of SACC,20 cases expressed PGAM1 at a high level,while 11 cases expressed at a low level.The PGAM1 expression was correlated with the pathological types,and was significantly higher in the solid pattern than the tubular and cribriform pattern(P<0.05).The expression of PGAM1 was not correlated with gender,age or tumor location(P>0.05).2?PGAM1 si RNA transfection specifically down-regulated the PGAM1 protein in SACC-LM cells.The downregulation of PGAM1 had no effect on the proliferation capability and cell cycle of SACC-LM cells(P>0.05),while cells migration and cell invasion were inhibited significatantly after PGAM1 was down-regulated(P<0.05).The expression of MMP-2 and MMP-9 was also down-regulated after PGAM1 was inhibited(P<0.05).Conclusions:1?The expression of PGAM1 was associated with pathological types of SACC;2?Down-regulation of PGAM1 can significantly inhibit the migration and invasion of adenoid cystic carcinoma...
