ObjectiveTo observe the effects of total flavonoids from Hypericum attenuatum Choisy on action potential of arrhythmic cardiomyocytes in rats induced by aconitine,and to explore the ion channel mechanism.Methods1.To observe the effects of total flavonoids from Hypericum attenuatum Choisy on APA.APD90,APD50 of the model cells using the whole cell patch clamp technique,and to explore the recovery effects on model cells.2.Whole cell patch clamp technique was used to observe the effects of total flavonoids from Hypericum attenuatum Choisy on INa of the model cells about the peak current density,the curve of I-V,the steady-state activation curve,the steady-state inactivation curve,the inactivation recovery time,to discuss the recovery action on exceptional INa.3.Whole cell patch clamp technique was used to observe the effects of total flavonoids from Hypericum attenuatum Choisy on ICa,L of the model cells about the peak current density,the curve of I-V,the steady-state activation curve,the steady-state inactivation curve,the inactivation recovery time,to discuss the recovery action on exceptional Ica,L.4.Whole cell patch clamp technique was used to observe the effects of total flavonoids from Hypericum attenuatum Choisy on IK1 on the model cells about the peak current density and the curve of I-V,to discuss the recovery action on exceptional IK1.Results1.The total flavonoids from Hypericum attenuatum Choisy(80?g/mL)can significantly decrease the APA of the model cells,and shorten the APD90 and APD50 significantly.2.Total flavonoids from Hypericum attenuatum Choisy(80?g/mL)have a significant inhibitory effect on INa peak current density on different level of depolarization on the model cells.Total flavonoids from Hypericum attenuatum Choisy(80?g/mL)make the INa I-V curve moved up,make the INa steady-state activation curve and the INa steady-state inactivation curve moved towards the direction of depolarization and prolong the recovery time of the INa3.Total flavonoids from Hypericum attenuatum Choisy(80?g/mL)have a significant inhibitory effect on ICa,L peak current density on different level of depolarization on the model cells.Total flavonoids from Hypericum attenuatum Choisy(80?g/mL)make the ICa,L I-V curve moved up,make the ICa,L steady-state activation curve and the ICa,L steady-state inactivation curve moved towards the direction of depolarization and prolong the recovery time of the ICa,L.4.Total flavonoids from Hypericum attenuatum Choisy(80?g/mL)make the IK1 peak current density of the model cells increased,and the IK1 I-V curve moved down.Conclusions1.The total flavonoids from Hypericum attenuatum Choisy(80 ?g/mL)can significantly decrease the APA and shorten the APD90 and APD50 of the model cells.This explains that the total flavonoids from Hypericum attenuatum Choisy(80 ?g/mL)can recover the action potential of arrhythmic cardiomyocytes in rats.2.The experimental results point out that total flavonoids from Hypericum attenuatum Choisy(80 ?g/mL)have a good healing effect on INa of arrhythmic cardiomyocytes in rats and its antiarrhythmic mechanism may be related to inhibiting fast sodium channel current.3.The experimental results point out that total flavonoids from Hypericum attenuatum Choisy(80 ?g/mL)have a good healing effect on ICa,L of arrhythmic cardiomyocytes in rats and its antiarrhythmic mechanism may be related to the inhibition of L-type calcium channel current.4.The experimental results point out that total flavonoids from Hypericum attenuatum Choisy(80 ?g/mL)have a good healing effect on IK1 of arrhythmic cardiomyocytes in rats and its antiarrhythmic mechanism may be related to enhancing inward rectifier potassium channel current.
