Objective:To discuss the mechanism by observing therapeutic action about the changes of behavior and the cerebral contents of 5-HT,5-HIAA,DA and NE which exist in depressed model rat.Methods:50 wistar rats were divided randomly into 5 groups:blank group,model group,group of Hypericum attenuatum Choisy. Low dose,group of Hypericum attenuatum Choisy. high dose and control group.Besides blank group,later 4 groups were made through cutting off water 24 hours,cutting off food 24 hours,baking at 40℃.swimming in the icy water,electric shocking the plantar surfaces of toe,infibulating tail,reversing day and night.After the chronic unpredictable stresses exerted on each rat excluding the control ones for 21 days.Day 8,treating group:one,Hypericum attenuatum Choisy was given according to 2.88 g/kg,the other 0.72 g/kg.control group was given hydrochloric acid fluoxetine according to 1.8mg/kg.Observing the changes of weight,detecting the contents of 5-HT,5-HIAA,DA and NE which exist in depressed model rat after 21 days.Result:1. The changes of weight show:Day 7,compared with the blank group,the weight of depression group has a significant difference(p<0.05), the gained weight of depression group has a very significant difference(p<0.01).Day 21,compared with model group, drug group weights were statistically significant.control group gained weight has a significant difference(p<0.05), neither low dose nor high-dose group has a statisticant difference, but they have certain effect on rats'weight gain.2. Rats activity show:Day 21,compared with model of rats,drug group level activities and vertical activities significantly increased (p<0.05), Hypericum attenuatum Choisy.can obviously improve the depression,reduce rat activity.3. The consumption of saccharu water show:Day 7,the level of depression group obviously lower than blank group(p<0.01),and continuing to 21 days.Day 21,compared with model group,the level of drug group has a very significant difference(p<0.01). Hypericum attenuatum Choisy.can reversal decrease the sucrose preference of chronic mild unpredictable stress rats.4. Variation of 5-HT,5-HI AA,D A,NE in content show:Day 21,model group of the corresponding was significantly lower than blank group.Compared with model group,high dose group of Hypericum attenuatum Choisy.and control group of 5-HT content increased significantly (p<0.01).High dose and control group of 5-HIAA content increased significantly (p<0.05). Low dose, high dose group and control group of DA content were not significantly increased (p>0.05). High dose and control group were obviously improved (p<0.05).Conclusions:1. Weight, saccharu water consumption, activity has diminished objectively after the chronic unpredictable stress,which is the same with behavior of depression。CUMS depression model provided the copy of success. Hypericum attenuatum Choisy.for depression rats not significant weight gain;Drug group can make depression rat sugar water consumption and activities that number increase. Experiments show that Hypericum attenuatum Choisy.can improve depression in a certain extent rats'depression performance.2. Levels of 5-HT,5-HIAA and NE in hippocampus of chronic unpredictable stress rat decrease, which is the same with monoamine transmitters viewer. High group can promote the level of 5-HT, 5-HIAA and NE in hippocampus of model rats, which may be one of the mechanisms of its anti-depressive action.
