Objective: Competition and training is an important part of competitive sports.The aim of competitive sports is to continuously tap the potential of the human body,to maximize the ability of athletes to compete and to create excellent athletic achievements.With the competition of modern competitive sports becoming more and more fierce,the intensity of competition and training is close to the limit of human body,which will bring different degrees of fatigue to the body.How to Resume Quickly,ensure the continuity and efficiency of the training and Competition,has been the goal of the majority of coaches and athletes.Acupuncture therapy has the advantages of less side effects and convenience in the recovery of human function after exercise,and will not produce stimulants and other issues.It is worth studying and promoting.But in the past,the research on acupuncture therapy mainly focused on the mechanism of a certain substance or several substances in a certain way,which could not reflect the overall effect of acupuncture therapy on the body.The metabolomics is to analyze the relationship between the molecular set and its function by measuring the changes in the concentration and proportion of various metabolites in the body to reflect the state of the body.It focuses on the body after a series of changes in metabolites of holistic change,rather than in the body by various external conditions to stimulate the change of a substance or metabolic pathways details,which allows it to be more accurate and comprehensive reflection of the body is external stimuli and produce a series of stress reaction.The effects of acupuncture on fatigue by using metabolomics are helpful to understand fatigue generation and the mechanism of acupuncture.Experiment I: Study on urine NMR metabolomics features of middle—distance runners after 800 m running and acupunctureMethods: In this study,18 Middle-distance athletes were selected from the Shanghai sports academy,and all of them were healthy and divided into experimental group(EA: 9): 800 m running + acupuncture,control group(ER: 9): 800 m running + natural recovery.Acupuncture points of choice "Zusanli,"Shenshu","Weizhong","Guan Yuan".Three sampling times were: 45 minutes before exercise,30 minutes after exercise,30 minutes after acupuncture.Urine was measured on a superconducting Fourier transform NMR spectrometer using the cpmgpr1 d pulse sequence and the signal was converted to an NMR spectrum by 32 k Fourier transform.The map folder into the Second Military Medical University test results reached more than 90% of the individual's best results,that is,the measured results were more than [100+(100-90)] * the individual best result /100,which was in line with the experimental requirements.(2)The 800 M fatigue scale shows that the athletes have fatigue after the 800 M run;The average heart rate immediately after the 800 M run was 202.5 / min,indicating that the athlete was highly aggressive during the 800 M run.(3)The analysis of 800 pre-run and post-NMR metabolic multivariate analysis indicators showed that there was a significant distinction between the metabolic features of 800 M pre-run and post-run urine samples.(4)800 run metabolic pathways involved in: based on the 800 m run before and after differentiation metabolic substances analysis found that the 800 m run metabolic pathways are involved in the metabolism of creatine,metabolism of amino acids,glucose metabolism,the Krebs cycle,purine nucleotide metabolism,tyrosine metabolism,etc.(5)The analysis of fatigue scale after acupuncture showed that there was no obvious difference between the two groups,indicating that the acupuncture effect was not obvious.Heart rate analysis after acupuncture showed significant differences between experimental group and control group.(6)After acupuncture,NMR metabolic multivariate analysis showed that there was no significant difference between the urine samples of the post-acupuncture experimental group and the control group,indicating that the acupuncture effect was not significant.Conclusions:(1)The causes of fatigue caused by changes in the related markers in the metabolic pathway involved in 800 m run are: energy consumption,large amount of lactic acid accumulation,oxidative stress and neural inhibition.(2)after the analysis of metabolites related to metabolites after 800 m running,it was found that the contribution of aerobic oxidation system to the energy supply was relatively large.(3)based on nuclear magnetic resonance(NMR)of urine metabonomics technology have non-invasive,simple processing,high sensitivity advantages,can provide new study idea for sports training,provide guidance for sports training.(4)after a 30-minute rest after the 800 m run,the metabolism of the body has basically recovered to the pre-run state.(5)the recovery effect of acupuncture treatment group was worse than that of natural rest group,and the reason was probably the two-way regulating effect of acupuncture.Experiment II: Study on urine NMR metabolomics characteristics of middle—distance runners after heavy-load training class and acupunctureMethods:In this study,14 Middle-distance athletes were selected from the Shanghai sports academy,and all of them were healthy and divided into experimental group(TA: 7): heavy load training + acupuncture,the control group(TR: 7): heavy load training + natural recovery.Acupuncture points of choice "Zusanli","Shenshu","DaZhui","Guan Yuan".Three sampling times were: 45 minutes before exercise,35 minutes after exercise,and 30 minutes after acupuncture.Multivariate NMR metabolic analysis is the same as experiment 1.Results:(1)The analysis of fatigue scale after heavy load training shows that athletes have fatigue after heavy load training;The average heart rate of the heavy load training class was 172.4 / min,indicating that the athlete's training load was relatively large.(2)Before and after heavy load training,NMR metabolic multivariate analysis showed that there was a clear distinction between urine samples before and after training.(3)involved in the metabolic pathway of heavy load training: through to the heavy load training before and after differentiation metabolic substances analysis found that the 800 m run metabolic pathways are involved in fatty acid metabolism,muscle acid metabolism,amino acid metabolism,sugar metabolism,Krebs cycle,purine nucleotide metabolism,methane,etc.(4)The analysis of fatigue scale after acupuncture shows that there are obvious differences between the two,and the fatigue recovery effect of acupuncture group is better.After acupuncture,the heart rate of the experimental group was significantly lower than that of the control group.Compared with the quiet heart rate,the heart rate of the acupuncture group was closer.(5)After acupuncture,NMR metabolic multivariate analysis showed that there was a significant difference between the urine samples of the post-acupuncture experimental group and the control group.(6)involved in the metabolic pathways of the acupuncture: through the acupuncture group and the control group after differentiation metabolic substances analysis found that after acupuncture involves the metabolic pathways of amino acid metabolism,sugar metabolism,TCA cycle,methane metabolism,purine nucleotide metabolism,metabolism of creatine,choline metabolism,etc.Conclusions:(1)The causes of fatigue due to the changes of related markers in the metabolic pathway involved in the heavy load training course are: energy consumption,large amount of lactic acid accumulation,oxidative stress,etc.(2)the athletes of this training course present the characteristics of anaerobic,aerobic and aerobic metabolism,which is in line with the characteristics of modern middle-distance race.(3)the orpiment purine(Hyx)and N-oxide trimethylamine(TMAO)in heavy load training and the 800 m run are significant sexual differences,human movement metabolic regulation may be important biological markers.(4)acupuncture related acupuncture points can help to relieve the acute exercise-induced fatigue caused by long time and large intensity.(5)the main influence of acupoint on acute exercise fatigue acupuncture is the energy metabolism pathway.(6)the mechanism of action of acupuncture related points to alleviate fatigue are: promote the synthesis of energy material,resisting oxidative stress,relieve brain fatigue(inhibition),protect cells,maintaining homeostasis.(7)the targeted target of acupuncture may be various enzymes on the metabolic pathway.(8)using NMR metabolomics to study acupuncture and moxibustion,it is helpful for qualitative,quantitative and objective evaluation of acupuncture effect.
