Objective:We established a rat model of chronic fatigue syndrome in this study.The metabolic pathway of CFS was screened by metabolomics research method,and the metabolic pathways of CFS were screened by the urine differential metabolites in quiet state and exercise intervention to provide experimental basis for further study of human CFS.At the same time,we explore the effect of exercise intervention on the rehabilitation of CFS rats,and provide a feasible treatment for the rehabilitation of CFS.Methods:(1)CFS model was established:the model of chronic fatigue syndrome was established by multi-factor induced compound modeling method using tethered ?sleep disturbance + crowded + treadmill.(2)Evaluation of CFS model:The effects of drinking water,body weight change,physical characteristics,water maze test results,open field test results and tail-tail test results were observed and recorded in this group.We use these to evaluate the success of the model and the healing effect of the treatment period.(3)Detection of urine differences and screening of metabolic pathways:To collect the urine of each group before and after exercise and metabolomics analysis,the use of GC-TOF-MS metabolomics technology to find the endogenous CFS rats Metabolites,using the multivariate statistical analysis of principal component analysis and orthogonal partial least squares to find potential differential metabolites.Met-PA databases were used to integrate the associations between different metabolites,construct the target path,analyze the corresponding Metabolic pathway.(4)The development of CFS exercise regimen in rats:After the rats were successfully modeled,the rats in the model group were randomly divided into two groups:one group was the exercise group and one group was the natural recovery group.The rats in the exercise group,the natural recovery group and the resting group had the same living environment and feeding conditions,and were fed into 40cm × 30cm × 20cm feeding cage respectively.The diet and drinking water were free.Rats in the exercise group were exercised with moderate intensity swimming at 7 pm daily.Swimming environment for the diameter of 150cm round pool,water depth of about 50cm,water temperature 33-36 ?.Exercise time is 60min/day.Treatment lasted for four weeks.(5)Evaluation of CFS exercise rehabilitation in rats:During the course of aerobic exercise rehabilitation,the body color,body shape,mood and mental state of rats were observed and recorded every day.Rats were weighed once every two days.The daily dietary and drinking water were recorded at a fixed time every day for weighing and weighing the remaining feed and drinking water.The rats were divided into two groups at the end of the second week and the fourth week,and the data were analyzed statistically.Results:(1)Test results of CFS rat model:Compared with the quiet group,the rats in the model group were divided into two groups:the drinking water,the weight,the general condition,the Morris water maze test,the open field experiment and the tail test And the rats in the model group were significantly different.The learning and memory ability and the ability of exploration were reduced,the avoidance and anxiety,and the depression were higher.Chronic fatigue syndrome rats were successfully modeled.(2)Test results of metabolomics:quiet state CFS rats than normal rats to change the metabolites are mainly ?-ketoglutarate,gluconic acid,galacturonic acid,indole-3-acetic acid,4-Hydroxybenzoic acid,3-hydroxyphenylacetic acid,inosine,malonic acid,21-hydroxypregnenolone,3-hydroxypropionic acid,and metabolite content of the trend are reduced.(3)The content of fumaric acid,dimethyl fumarate,malic acid and palmitic acid increased in CFS rats compared with normal rats after one-time exercise;malonic acid,a-ketoglutaric acid,tartaric acid,Gluconate,sphingosine,21-hydroxypregnenolone content decreased.There were three metabolic pathways in CFS rats and normal rats,which were tricarboxylic acid cycles,alanine,aspartic acid and glutamic acid metabolism,and the synthesis of steroid hormones.There were four metabolic pathways in CFS rats and normal rats after exercise,which were tricarboxylic acid cycles.Alanine,aspartic acid,glutamic acid metabolism,synthesis of steroid hormones,sphingolipids metabolism.(4)Rehabilitation result of CFS rats:After 14 days of exercise rehabilitation treatment,CFS rats' diet water,weight,open field test results,water maze test results,tail test results significantly improved and returned to normal.There was no significant difference between the two groups.Conclusion:(1)The complex symptoms of CFS rats may be due to theabnormalof TCA cycle;alanine,aspartic and glutamic acid metabolism;Steroid hormone synthesis.It can be found an extra pathway which iasphingolipid metabolic also abnormal after exercise.So disease pathogenesis can be more in-depth detection when movement as a means of disease detection.(2)After administration of aerobic exercise in rats,the symptoms of CFS rats can be significantly improved.It can relieve the fatigue state of rats and improve the spatial memory and learning ability,the ability of independent exploration and the activity of autonomic activity of CFS rats.
