Objective To understand how Shen Qi compound recipe (SQCR) works, which with the function of tonifying Qi and Yin and promoting blood circulation, in treating diabetes mellitus macroangiopathy in GK rats on low grade inflammation and endothelial cell apoptosis, and to find out the possible pathway. To unreveal the micromechanism of treating diabetes mellitus macroangiopathy, so as to provide the theory for tonifying Qi and Yin and promoting blood circulation for treating diabetes mellitus macroangiopathy.Methods 73 GK male rats were randomized to four groups:18 in GK group(drenched normal saline 5ml·kg-1·d-1),19 in model group (drenched normal saline 5ml·kg-1·d-1),18 in atorvastatin group(drenched atorvastatin calcium suspension 1.6mg·kg-1·d-1),18 in SQCR group(drenched SQCR suspl 1.44g·kg-1·d-1),18 SPF Wistar rats were used as the control group(drenched normal saline 5ml·kg-1·d-1). The N?-nitro-L-arginine methyl ester (L-NAME) 0.1mg·ml-1·d-1 was given in the drinking water for atorvastatin, SQCR and model groups to build the diabetes mellitus macroangiopathy model:GK rats were fed with high fat diet, while Wistar rats with general diet. Treating while setting up model. In the experimental periods, general status were observed as well as the amount of water drinking, food intake, blood glucose and body weight measured. At the end, the blood glucose, blood fat, blood secum C reactive protein (CRP) and TNF-?were detected. The caspase-3 expression of abdominal aorta EC and COX-2 mRNA expression in the abdominal aorta vessel wall were detected, the apoptosis of thoracic aorta EC and the akt, phospho-Akt(Ser473), eNOS, phospho-eNOS (Serl 177) protein expression of thoracic aorta were detected.Results The random blood glucose and body weight were no statistical difference among GK rats, except comparing GK rats with Wistar rats (P<0.01). The amount of water drinking between two experimental groups and model group were statistically different (P<0.01) from the 3rd week of the experiment. The amount of water drinking between SQCR group and atorvastatin group were statistically different (P<0.05) from the 4th week of the experiment. So SQCR could decrease the amount of water drinking. The amount of food intake between two experimental groups and model group were statistically different (P<0.05) from the 5th week of the experiment. The body weight was increased and fasting blood glucose was decreased in SQCR group compared with model group at the end of experiment(P<0.01 or P<0.05). The caspase-3, COX-2 mRNA expression of aorta and blood serum total cholesterol, triglyceride, CRP, TNF-a in model group were all higher than other groups(P<0.05 or P<0.01). SQCR could reduce blood serum total cholesterol, triglyceride, CRP and TNF-a, lessening inflammatory reaction, inhibiting caspase-3 and COX-2 mRNA expression, suppressing apoptosis of vascular endothelial cell(VEC). SQCR could also restrain phospho-Akt (Ser473), phospho-eNOS (Ser1177) protein expression in aorta.Conclusions SQCR could decrease the amount of water drinking while increasing body weight of model rats, improving glycolipid metabolism, so it may play the role of anti-inflammation and reducing macrovascular complications in GK rats, which may be realized through suppressing apoptosis of VEC, reducing blood serum CRP, TNF-a, inhibited caspase-3 and COX-2 mRNA expression, restrain the phospho-Akt (Ser473)?phospho-eNOS (Ser1177) protein expression in aorta, which could inhibit neovascularization and delay atherosclerosis(AS).
