Objective To clarify the effects and its mechanisms of ShenQi compound recipe (SQCR) with the function of tonifying Qi and Yin and promoting blood circulation to dissipate stasis and representing the fundamental methods treating type 2 diabetes mellitus (T2DM) on inflammation and macrovascular complication in GK rats so as to provide the theory base to apply Traditional Chinese Medicine in clinic. Methods Copied T2DM model with low grade inflammation and macrovascular complication by injecting Nω-nitro-L-arginine methyl ester (L-NAME) intra-peritoneally and high fat diet, animals were randomized to normal Wistar control, model, ramipril (positive control), ShenQi compound recipe low dosage and high dosage (SQCRL and SQCRH) groups and administrated water, water, ramipril, SQCRL and SQCRH respectively for 32 days. In the experiment periods, observed general status and measured the amount of water drinking, food intake and body weight. At the end, determined serum glycation serum protein (GSP) by fructosamine assay, C-reactive protein (CRP), monocyte chemoattractant protein (MCP)-l and adiponectin by ELISA, tumour necrosis factor(TNF)-a by radioimmunoassay. The mRNA expression of peroxisome proliferator activated receptor (PPAR) y and adiponectin in white adipose tissue and PPARy, MCP-1 and nuclear factor (NF)-kB p65 in aorta by real time RT-PCR, NF-kB activation using immunohistochemistry staining and observed thoracic aorta pathomorphology under light microscope. Results The general status, the amount of food intake and GSP in SQCR and ramipril groups were no significance compared with model group, but at the end of experiment, the amount of water drinking was decreased in SQCRL and ramipril groups and the body weight was gained in SQCRH compared with model group (P<0.01). Concentrations of CRP and TNF-a in serum were all decreased in SQCRL, SQCRH and ramipril groups compared with model group (P<0.05 or P<0.01).
