| Allium Sativum L. is a plant belonging to family of lily , genera of shallot . Its bulb is named garlic . Allicin is some active organic sulfide which is come into being from Alliin by catalyzing of Alliinase when the fresh bulb is crashing . Allicin has a broad-spectrum function for anti-bacteria and anti- epiphyte ; can clear away free radical from body , protecting cells from cancerating and protecting blood vessel from cerebrovascular disease ; also can enhance organism's immune ability , and so on . Allicin has a lot of effect on pharmacological aspects and extensive curative effects , and its clinic application is more and more popular . Allicin is a promising crude drug . Allicin (diallyl -sulfinothioicacide ) is very active , and it is unstable when it is heated , exposing in the air or dissolved in organic solvent ; and it is easy to be decomposed into little active organic sulfide . So it is a very difficult thing for people to extract Allicin effectively for a long time , and its preservation is also a very challengeable problem . The methods of extracting rough Allcin ( garlic oil ) can be classified three groups : distilled by vapor after decomposition by Alliinase , extracted by organic solvent and extracted by super-clinic carbon dioxide . There is little impurity in garlic oil extracted by distilled by vapor after decomposition by Alliinase but little Allicin , either , otherwise , there is a lot of needless substance in garlic oil extracted by super-clinic carbon dioxide and with a boring subsequent purification . In this thesis , method of extracted by aether after decomposition by Alliinase was adopted to obtain rough product ( garlic oil ) . The extraction by aether was carried through following primary removing impurity after bulb crashed and decomposed by Alliinase . It was found that the rough product can easily be solved by aether . In this experiment , it was begun with amelioration of method of fix-sulfur , and then confirmed the process of operation of ameliorated method of fix-sulfur . The feasibility of the ameliorated method of fix-sulfur is proved by experiment of BaSO4 and blank experiment of rational filter paper . Subsequently , the experiment of choice of extraction solvent after decomposition by Alliinase was carried on , and it was found that aether is more favorable to extract garlic than that of Ethyl acetae through their comparison on time of extracting and volume of solvent using . And then , aether was chosen to be the solvent to extract garlic after decomposition by Alliinase . In the following experiments , we started to examine the mono-factor , such as temperature , pH value of solution , the volume of water , time during decomposing by alliinase , pH value of solution before centrifugation , and so on . and the range of the yield of garlic organic sulfide was gained . Finally , two group of orthogonal experiment which we want to find out the noticeable factors and the orthogonal experiment which we want to confirm the optimum conditions of extraction were carried on . As a result , the factors of temperature , pH value of solution , the volume of water , time during decomposing by alliinase , pH value of solution before centrifugation were noticeable , which influenced the yield of garlic organic sulfide . The optimum operation condition was confirmed through the orthogonal experiment : A1B2C2D2H4 , i.e. maintaining the following conditions : the temperature during decomposing by alliinase , 25℃ ；the pH value of solution during decomposing by alliinase , 7.0 ; the volume of water , 100 ml ; the time during decomposing by alliinase , 60min ; pH value of solution before centrifugation , 3.2 ; and it was proved that the yield of garlic organic sulfide was 0.188% under the optimum condition .
