In this paper, octanol-water system was adopted to study the distribution of decoction in stomach and intestine. Analyzed the metal speciation and effective chemical composition of three Chinese traditional medicine: Chinese Angelica root, Astragalus root,Scutellaria root. By FAAS,HPLC and UV-Vis spectrophotometer. The different acidities of digestive site, the different composition of medicine and compatibility of medicines have greatly affective the species of metal elements and the effective chemical composition.In chapter one, the study of Chinese Angelica root, Astragalus root and Scutellaria root were summarized imply.In chapter two, the total concentration, octanol– and water–solubility concentration of seven elements in medicinal materials or decoctions under gastric and intestinal acidity, and extract percent of TEs in Chinese Angelica root and Astragalus root, were determined, respectively, by FAAS, analyzed and compared. Results indicate that the different acidities of digestive site, different composition of medicine, and the compatibility of medicines, have greatly affected the existing species of metal elements, the pharmacological, activity of coordinated metal complex in decoctions. The octanol-solubility concentration of seven elements is higher under intestinal acidity. The extract percent of seven elements is higher when compatibility of ChineseAngelica root between Astragalus root is 1:5.In chapter three, HPLC was used to establish the fingerprints of Chinese Angelica root, Astragalus root and DanggBueTang under different acidities, the data were analyzed and compared. There are eighteen effective chemical compositions in Chinese Angelica root, twenty two in Astragalus root. The quantity and concentration of effective chemical compositions are different under different acidities. The different acidities of digestive sites, different compatibility of medicines have greatly affected the species of the effective chemical composition.In chapter four, the speciation of eight elements in Scutellaria root, were investigated, the fingerprint of Scutellaria root under different acidity were established. The concentration of Baicalin, main validity component in Scutellaria root, is different under different acidities; we presume that Baicalin could be combined with other components and exist in decoction or human body in combinative speciation.In chapter five, the UV-Vis spectrum characteristic and antioxidant action of Baicalin were studied when combined with different metal elements. The results indicated that Baicalin can coordinate with metal elements easily under low acidity. the antioxidant action of Baicalin was improved by metal element. The antioxidant mechanism of Baicalin with different metal elements in different systems were also investigated.
