Water-soluble crude polysaccharide has been extracted from spores of Ganoderma Lucidum. Analysis indicated that it contained glucose (Glc) and galacose (Gal), and its molar ratio is 16.53:1.00. The crude polysaccharide has been deproteinizationed by the combined methods of enzyme and Sevage, fractionated and purified by Sepharose CL-6B gel filtration. Three fractions: GLP1,GLP2,GLP3 were got respectively. So GLP1 was got .By gel filtration on Sepharose CL-6B, HPLC and specific optical rotation measurement test. GLP1 was a single strait symmetry apex. The identification of purify implies that GLP1 was respectively uniform on molecular weight and polarity. GLP1 was a gray white substance. G.C. analysis indicted that GLP1 was only composed of Glc. Partial hydrolysis with acid and hydrolysis with fibrin enzyme, Periodate oxidation, Smith degradation, I.R., G.C., Methylation and 13C-NMR were used on the structure analysis of GLP1. Methylation production were hydrolyzed, deoxidize, then analyzed by G.C.-M.S. The results revealed that GLP1 was a kind of highly branched structure. The main chain was composed of (1→6) -linked-Glc, and (1→3)-linked-Glc residues were substituted at O-3, O-4. The results revealed the main chain of GLP1 is made up of (1→6)-linked- Glc as well as substituted at 0-4 and 0-3. On average, there are 3 branches among 10 main chain residues. In vitro antitumor activities were investigated by MTT methods The results revealed that GLP1 had no direct killing action for hela cells (P>0.05). The anti-tumor action of GLP1 in mice was mediated by the corpus immune system.
