A water soluble crude polysaccharide has been isolated from the root of Codonopsis pilosula by boiling-water extraction and ethanol precipitation. The crude polysaccharide was analysed by G.C. It is consist of Glc,Gal,Xyl,Rha,Ara,GalA,the molar ratio is 102.7:7.9:2.2:1.8:1.1:1.0。 The crude polysaccharide was deprotinized by Sevage method and freezed-melted repeatedly, fractionated by ultrafiltration and gel chromatography Sepharose on G-75 column, so CPPS3 was obtained. The results of GC analysis revealed the monosaccharide of CPPS3 is made up of Gal,Ara and Rha with the molarity rate of 1.13:1.12:1,as well as a few of Glc. Its homogeneity was proved by HPLC and specific optical rotation measurement et, al. It was proved to be homogeneous polysaccharide on molecular weight and polarity as well. A colorimetric tetrazolium MTT method was used for the determination of anti-tumor activity evaluation, but no sufficiently directly cytotoxic effect was detected. The totle sugar content of CPPS3 is 94.5%. Its average molecular weight is 106kd by means of HPLC. In the structure analysis of CPPS3,IR,G..C.,partial hydrolysis with acid, periodate oxidation, Smith degradation, IR. , G.C., (13)CNMR spectroscopy, methylation and GC-MS analysis were the principal methods used. The results revealed the main chain of CPPS3 is made up of (1→3)-linked- Rha and (1→3)-linked- Gal, as well as substituted at 0-2. On average, there are 2 branches among 5 main chain residues. The side chain is composed of (-1)Gal4- , (-1)Ara5-, (-1)Rha2- and (-1)Rha3-; The nonreduced end is composed of Araf.
