Traditional Chinese Medical prescription as the primary form in clinic of Chinese Medicine plays an important role in developing Chinese Patent Medicine and new Traditional Chinese Medicine. Complex Astragalus mainly contained Astragalus and Glycyrrhiza was demonstrated to behave obvious hypoglycemic activity, and n-butyl alcohol extraction of its aqueous extract was proved to be efficient component by pharmacological activity screen. To illuminate the therapeutic mechanism of Complex Astragalus, chemical constituents of its effective component have been isolated for the first time by means of column chromatography, preparative thin layer chromatography and recrystallization etc. From n-butyl alcohol extraction and diethyl ether extraction of its aqueous extract, more than 16 compounds have been isolated. 10 compounds were obtained from n-butyl alcohol fraction and 6 from diethyl ether fraction. In combination with the data from literature, the structures of those compounds have been elucidated on the basis of spectroscopic methods (IR, UV, FAB-MS, 1D/2D NMR). All these compounds were identified as xanthones, flavonoids, triterpenoid saponins. Five xanthones were found for the first time in Complex Astragalus and detected in the ethanol extraction of the aqueous extract by HPLC.Codonopsis pilosula (Franch.) Nannf. is a commonly used Chinese medicine. It has been employed for treatment of dyspepsia, poor appetite, fatigue and psychoneurosis, etc. In this thesis, GC-MS techniques were used in analyzing the essential oils of the C. pilosula. A diversity of biological activities for Polysaccharides in C. pilosula has been demonstrated by pharmacological researches, including antitumor, antiviral, antioxidative, antiulcer activities and immunomodulatory effect. Our intent of the present investigation focused on optimizing the preparation of the polysaccharides from C. pilosula and creating a simple procedure to purify the polysaccharides. An optimum extraction of polysaccharides from C. pilosula has been established by orthogonal test. After purifying the crude polysaccharides, a polysaccharide CPPS-I that possessed high content has been prepared by membrane separation and evaluated by IR, UV, GC-MS, XRD, SEM, AFM, 1D NMR etc.This thesis will provide more chemical evidences for researching the Traditional Chinese Medicine and exploiting the polysaccharides.
