Polyamidoamine star-burst dendrimers (PAMAM) were bonded to the interior of silica capillaries through alkylation reaction with 3-aminopropyltriethoxysilane, the Michael addition reaction with methylacrylate and animation reaction with ethylenediaine. The open-tube capillary electrochromatography columns (OCEC) modified with different generation of PAMAM were obtained by repeating the reaction steps. The obtained columns were characterized by CEC process. The results showed that the electroosmotic flow (EOF) gradually reduced with increasing PAMAM generation. Different generation of PAMAM was synthesized on the silicon plate with same reaction condition. The obtained silicon plates were characterized by scanning electron micrograph and X-ray photoelectron spectroscopy. It was demonstrated that the PAMAM have been bond to the inner capillary.The separation performance of OCEC modified with different generation of PAMAM was studied. Alanine(Ala) and proline(Pro) were selected as separation model targets. The results indicate that the resolution of alanine(Ala) and proline(Pro) was gradually improved with the amount of PAMAM generation increasing. By open-tubular capillary electrochromatography column modified with 3 generation PAMAM, the baseline separation of the alanine and proline can be obtained.
