Tea protein(TP) was extracted from depolyphenols tea residues with a mixed solutionsystem which was optimized as: the NaOH concentration 0.06mol/L, the extractiontemperature 90℃, the extraction time 40 min. and tea :water ratio 1:40. Under this conditionthe maximum extracting rate of TP was 56.4%. After the solution was decolored with activecarbon and adjusted to pH4.5 the crude TP was prepared. The trial of HFR clearanceindicated that the IC50 value of raw and decolored TP solution was 5.89mg/ml and 11.5mg/mlrespectively.The molecular weight range of TP was estimated with Shodew PROTEIN KW-802.5 to5000~1000,000Da and TP-3-2 whose IC50 value was 20.3mg/ml was obtained throughSephedex G-200 and DEAE(FF). By the electrophoresis the molecular weight of TP-3-2 wasappraised to 28800Da.The possibility of adding tea residues in the Ganoderma lucidum medium was studied.The optimum adding content was 1.0% and the optimum fermentation time was 105~110h.Under this condition 4.46g/L polysaccharides were obtained. At the same time there were fewchanges in amino acid content of hyphal pellets. All results above indicated that there was noeffect of adding 1.0% tea residues on Ganoderma lucidum growth.It was confirmed that all peptides provided with the ability of HFR clearance, amongwhich the intracellular peptides adding tea residues had the highest ability of IC50 0.31mg/ml.Through RP-HPLC some changes in its component were observed. The result of amino acidcontent indicated that compared with intracellular peptides without adding tea residues Gluand Pro increased by 78.0% and 40.1% while Arg and Lys reduced by 27.4% and 78.5%respectively.The results in vivo confirmed that the TP had remarkable effect on deceasing the bloodlipid of hyperlipidemia and preventing atherosclerosis and coronary heart disease while noeffect on anti-senility. However the Ganoderma lucidum ferment liquid whether adding tearesidues or not had the function of anti-senility that MAD in old mice decreased by 39% and27% and SOD increased by 16% and 14% respectively.
