Ganoderma Lucidum is a kind of eumycete medicine with full of nutrition and high value of health protection. Modern science demonstrate that which has the effect of resisting tumor,resisting anti-aging, getting rid of oxygen free radical, promoting blood flow,resisting thrombus and protecting liver. At present, it has attracted the concern of many experts of pharmaceutical and health industry. This paper mainly investigated the extraction and optimization of the process of water-soluble and ethanol-soluble protein form Ganoderma lucidum, the detection and analysis of physicochemical properties of enzyme reaction of the protein. It is significance to the development of the new drug and the functional health products.The main contents and some conclusions of the thesis are as follows:1. The extraction technology of water extraction protein from Ganoderma lucidumAccording to the response surface mathematical model which established carries on the parameter optimization analysis, obtains the highest parameter condition of water-soluble protein extraction: material liquid ratio is 34.42mL/g, soaks temperature is 26.56℃, soaks time is 8.87h, and pH is 8.89. In this optimization craft, the soluble protein extraction rate's predicted value is 82.59mg/g. In the confirmation experiment, the extraction rate of the water-soluble protein is actually 81.35mg/g, which demonstrate there is no significant difference in predicted and actual values. The influence factors on the extraction rate of water soluble protein influence is as follows: pH﹥extraction temperature﹥extraction time﹥material liquid ratio, and the influence induced by pH value to the experiments is extremely remarkable.2. The extraction technology of ethanol-soluble protein from Ganoderma lucidumEthanol-soluble protein was extracted form Ganoderma lucidum. The effects of extraction time, extraction temperature, pH, material-liquid ratio and extraction frequency on the yield of ethanol-soluble protein were studied with single factor and orthogonal tests. The single factor experimental result indicated that the best casft is obtained when material-liquid ratio was 1:40, extration time was 2 h. The effects of factors on yield of ethanol-soluble protein from Ganoderma lucidum is as follows: pH > material-liquid ratio > extraction temperature > concentration of ethanol. The optimal conditions for extractions as follows: extraction temperature is 65℃, extration time is 8 h, material-liquid ratio is 1:40, and the extraction times are 2. In this optimizing condition. In validation experiments, the actual measured alcohol-soluble protein extraction rate is 10.82%.3. The properties research of enzyme hydrolysis of Ganoderma lucidum proteinThrough the analysis by means of SDS-polyacrylamide gel electrophoresis, the composition and content of enzymatic amino acid have been qualitative and quantitative detected. The solubility, antioxidant, physical and chemical properties etc were investigated, and the antioxidant of ethanol-soluble protein and Vc were compared with validation under the same conditions. These results indicate that the enzyme hydrolysis of ethanol-soluble protein has significant anti-oxidation performance with good efficiency, and the ability of the hydroxyl radical and reducing power of enzyme hydrolysis of ethanol-soluble protein have been enhanced. The reducing power of Ganoderma lucidum protein with the concentration 4.2mg/mL approximately quite to the reducing power of Vc with the concentration 0.05mg/mL. As the fungus protein concentration increased, the ability to clear the ? OH radical increased; when the fungus protein concentration is 0.28mg/mL, the scavenging rate of·OH is 19.22%. The relationship between the concentration of the fungus protein and the ability to clear the·OH can be described by logarithmic equations.
