It is very important to study the effective fraction (EF) for the modernization of Chinese Traditional Medicine (TCM). The EF includes the most effective components of TCM. Through the rational combination or application of EFs, the TCM is shown to be the characteristics of multi-component, multi-pathway and multi-target. Moreover, the clarification and enrichment of the bioactive components in EF are beneficial to the application of modern extraction, purification technologies and new agents. Thus, the quality of EF can be well controlled and get the criteria of safety, efficiency and good quality.There are many issues should be discussed in the research of EF, such as extraction, purification technology and quality control. In this study, Radix Paeoniae Rubra was chosen as the object for systematic studying the key problems of extraction, purification and quality control of EF.1. The optimization in extraction and purification of EFThe single factor and orthogonal design methods were employed to investigate the main factors including extraction solvents, extraction times, consumed time and volume of solvent. We applied macroporous resin (MR) to concentrate the active components and investigated some parameters of purification process including adsorption capacity of MR, initial sample concentration, elution solvents and flow rate, etc. An GC method was established for the analysis of 6 venenous organic solvents.2. The quality control of EFIn this study, several methods were developed to determine the total glucosides of paeony. Meanover, we established the HPLC and LC-MS methods for analysing the bioactive components in EF. The stability of preparation process was evaluated. It indicates that the integrated method was an overall, valid and objective method to control the quality of EF.
