Traditional Chinese Medicine (TCM) is a precious heritage of our China. After continuos clinical application and development in nearly 5000 years, it has not only been widely accepted by Chinese but also it now has remarkable influence in Europe, America and other Asia countries. The quality control of TCM is a very important task and this is the foundation of modernization of TCM. The quality control by chromatographic fingerprint is the hot spot and the trend in these years. So, quality control of TCM by chromatographic fingerprint was selected as the topic of this thesis.In this thesis, four fundamental problems (A,B,C and D) were carefully studied and some useful harvests have been got. The details of these problems and research results are listed as follows:A. How to systematically optimize the operational condition of binary RP-HPLC fingerprints? The first step of this optimization is to get the value of retention parameters of solutes, which were commonly achieved by linear regression of several isocratic retention values. However, for practical complex samples, this method is tedious and time consuming and is difficult to be applied. In chapter two of this thesis, a new numerical emulation algorithm was established to calculate retention parameters in RP-HPLC with several retention times under different linear or nonlinear binary gradient elution conditions and further predict the retention time under any other binary gradient conditions. A program was written according to this algorithm and nine solutes were used to test the program. The prediction results were excellent. The maximum relative error of predictedretention time was less than 0.45%. With the help of this program, we calculated the retention parameters of components in the chloroform extract of Swertia franchetiana. Further more, the HPLC condition of this extract were optimized with these retention parameters. The final optimization result is fairly good.B. How to calculate the similarity between two chromatographic fingerprints? How to finish this calculation automatically?In this paper, On the basis of vector angle expression, a new method was presented to evaluate the similarity. Further more, automatic peak matching between two chromatograms was realized according to three rules as follows:(1) Under the same test condition, the retention time difference of the same component in the two chromatographic fingerprints is less than a reasonable value.(2) Under the same test condition, peak elution order of the fingerprint is immobile.(3) Find the most similar peak match result when abide with (1) and (2).With the combination of automatic peak matching and vector angle expression, an algorithm was put forward to automatically calculate the similarity between two chromatographic fingerprints, and a software was written according to this algorithm. This algorithm will be described and discussed in chapter four of this thesis.C. Now, lots of fingerprints of many kinds of TCM have been accumulated. It is very useful to establish a fingerprint database to store these fingerprints and relative information. This fingerprint database can provide beneficial reference for the people who engage in quality control of TCM. This fingerprint database was design and built in this paper. The detail of the database will be discussed in chapter five of this thesis. .D. Retention parameter can also be utilized to discriminate different components. In chapter three, retention parameter was used to discriminate the xanthones in themethanol extract of Swertia franchetiana. It has been found that the retention parameters of xanthonoids are in specific region in the plot of Logkw vs S. Xanthonoids in Swertia. extract could be distinguished from other components by this feature. Further more, two different xanthones, free xanthonoids and xanthone glucosides, can also be discriminated by the retention parameters.
