Cortex Moutan are cortexs of root of Paeonia suffruticosa Andr., total glucosides in Cortex Moutan has anti-oxidative, immunomodulatory, anti-hepatic injury, cardioangiography effects. Fructus Crataegi are the fruits of Crataegus pinnatifida Bge., total flavonoids has obvious curative effecton diseases of heart and blood vessel. We studied on analysis of paeoniflorin and total glucosides in Cortex Moutan and separation-purification process of Total Glucosides, and on analysis of hyperoside and total flavonoids in Fructus Crataegi and separation-purification process of total flavonoids. The results are followings.1. Study on analysis of paeoniflorin and total glucosides in Cortex MoutanThe experiment determined the analysed method of content of paeoniflorin and total glucosides in Cortex Moutan by the HPLC with paeoniflorin and benzoic acid as control. Results showed the two methods had a good linear range, accuracy, stability, responsibility and recovery, and may serve as a way of quality stantard draving up.2. Study on separation-purification process of Total Glucosides in Cortex MoutanThe experiment was designed by orthogonal design and was evaluated by the content and elution ratio of total glucosides, and detemine the craft flow-sheet extracting the total glucosides: using 70%alcohol refluxing extraction. The optimum technological parameters of purification was detemined: the macroporous resin ZTC-1 was the best for adsorption and separation of the total glucosides;the adsorption ratio for total glucosides in Cortex Moutan on ZTC-1 resin was 6.73mg/g;the best amount of the sample purified by the resin was half of the adsorption;the eluting agent, 15% agent ethanol (7 times the volume of the resin);the eluting velocity0.4ml/cm2 ? min. ZTC-1 resin could be used to separate and purify the total glucosides from Cortex Moutan, the content of total glucoseides in the product is 53%, and the eluted ratio reaches to 60%, and the technological process had a better reproducibility and could be used in magnified produce.3. Study on analysis of hyperoside and total flavonoids from Fructus CrataegiThe experiment had determined the analysed methode of content of hyperoside in Fructus Crataegi by the HPLC. The content of the total flavonoids was analyzed through colored with aluminium nitrate-sodium nitrite, and in an ultraviolet-visible spectrophotometry with hyperoside as the control. The two methods had a good linear range, accuracy, stability, responsibility and recovery, and may serve as a way of quality stantard draving up.4. Study on separation-purification process of total flavonidsThe experiment was designed by orthogonal design and was evaluated by the content, and detemine the craft flow-sheet extracting the total flavonids from Fructus Crataegi: using 60%alcohol refluxing extraction. The suitable resin to purify the total flavones was detemined and to establish the optimum technological parameters of purification. The DlOl resin was the best material for adsorption and separation of the total flavonids by the following technological coefficient: the dynamic adsorption ratio for the total flavonids was 27.55mg/g;the eluting agent was 6BV 60%ethanol and eluting velocity was 3.0ml/min. The results showed that the DlOl resin could be used to refine the total flavonids from Fructus Crataegi, and the content of total flavonids in the extract is 54%, the eluted ratio is 60%, and the technological process had a better reproducibility and could be used in magnified produce.
