Fructus aurantii has been used as a Chinese medicine for a long history. It has the function of dispelling and eliminating the accumulation, reducing phlegm and dispersing the ruffian. Total flavonoids are the key active agents in Fructus aurantii. The research of the Pharmacodynamics shows that the total flavonids of Fructus aurantii have the bidirectional adjusting effect to the intestinal smooth muscles. It has low toxicity, large safty range and very distinctly officinal value.This thesis focused on the microwave-assisted and ultrasonic-assisted extraction processes of total flavonoids from Fructus aurantii. The influences on extraction efficiency of the total flavonoids, including Fructus aurantii grain diameter, solvent concentration, ratio of liquid to solid, ultrasonic/microwave power and extraction time, were analyzed. Based on the signal factor investigation, a polynomial quadratic model was set up by the principles of the central composite design and the Response surface analysis methodology (RSM) to optimize the extraction conditions.The optimal conditions of ultrasonic-assisted extraction of total flavonoids from Fructus aurantii was concluded as follows:Fructus aurantii grain diameter was60mesh, ethanol concentration was60%(v), ultrasonic power was300W, ultrasonic-assisted extraction time was30min and ratio of liquid to solid was15:1.Meanwhile the optimal conditions of microwave-assisted extraction of total flavonoids from Fructus aurantii was determined as follows:Fructus aurantii grain diameter was40mesh, ethanol concentration was60%(v), ratio of liquid to solid was10:1, microwave power was480W, microwave-assisted extraction was4min.The separation and purification process of the total flavonoids in Frutus aurantii with macroporous resin was studied. The static adsorption tests of the seven different kinds of macroporous resins were investigated to select the optimal macroporous resin for the purification of total flavonoids. It was found that the macroporous resin of model AB-8could purify the total flavonoids in Frutus aurantii more effectively. The recovery rate of the total flavonoids would be the highest if the sample concentration was0.408mg/ml, the sample flow rate was2BV/h, the sample pH was 4. When using6BV75%(volume fraction) ethanol as eluent at1BV/h speed, the desorption rate of total flavones could reach up to94.85%.A glass column (Φ3.0cm×60cm) packed with model AB-8macroporous resin was used for the total flavonoids purification from Fructus aurantii. The layer of macroporous resin was34cm height, namely1BV (column bed volume) of about240mL.the liquid sample of flavonoids purification increased from the original14.03%to42.54%. The separation process of the flavonoids from Frutus aurantii was also studied in this paper.
