8-methoxypsoralen ( 8-MOP ) and praeruptorin A ( Pd- I a ) in Peucedanum praeruptorum Dunn were identified by TLC with spot clear and reproducible. Quantitative assays were performed on the two active components of 8-MOP and Pd-I a by HPLC. Active components had good linearity with the range of 0.05 μ g~0.35 μ g(r=0.9996) and 0.2 μ g—1.8 μ g(r=0.9996) respectively. And the average recoveries of them were 98.0%(RSD=1.72%)and 98.3%(RSD=1.51%). The two methods can be used for the quality control.The effects of pressure, temperature, extraction time, the flow rate of CO2, the CO2 density, kinds of entrainer on the extractive results of Peucedanum praeruptorum Dunn were explored, at the same time , the curves of the influence factors were drawn. The optimum conditions for SFE- CO2 was selected with the aid of orthogonal design by using the content of 8-MOP , Pd- I a and the yield of extractor as quality control criteria. The results were as followed: pressure 25MPa, temperature 60℃, extraction time 3h, 95%alcohol as entrainer (1 : 1 w/v) and the rate flow of CO2 3.2L/h. Compared with the technology of traditional solvent extraction, SFE-CO2 of Peucedanum praeruptorum Dunn was proved to be efficient, simple and save the extraction time.
