Glutathione (GSH) is an activated tripeptide which has important physiological function. It iscondensated by glutamic, cysteine and glycine. It widely exists in the nature, the theanimal liver,theyeast and the plant embryo all has rich GSH, its content reaches as high as 100-1000mg/100g.. Invivo, GSH is the responses auxiliary base of many enzyme, It can eliminate free radical, detoxicate,accelerate the absorb of iron substrate and participate the traffic of amino acid, etc. It is not only widelyused in medicine,biochemistry and chemistry, but also used in making functional food as the idealchemical additive.Corn's embryo is specially big, its weight approximately is 10~13%in the whole grain. In the cornembryo, the protein content reaches about 18%-30%, the fat content is 36%-59%, and in the embryo thenutrition quality of the protein and the fat is extremely high, extremely has takes the food industry foodsource the development use value. Jilin province is one of the corn main production areas in China. Theyield and stocks of corn comes out top at home and broad every year, but because of lacking comprehensiveutilization, high technology, high additional value and market potentiality, the advantage of JiLin cornindustry can not exert sufficiently by far. In this thesis the corn corcule as a coproduct material is used toextract and depurate the tert functional food raw material-GSH with the purpose of providing the industryproduction technique. By this project, It can extend the corn industry chain utilitily, cut down the cost of thecorn alcohol and starchy and the corn high additional value enterprise can be promoted sigiificantly. It canpromote the development of the corn economy in JiLin province and develop the palae-industry depot innortheast.The topic originates of the research is selected from《Study on extractive technique and applicationof free radical counteractant》of JinLin province science technology department.Manily reaches on theextraction and depuration technique of GSH in the corn corcule and has obtained below the result:1. In the corn embryo, the fat content reaches as high as about 58%, which will effect the extractionand depuration of GSH, so we must extract it thoroughly. Compared to some different methods, we selectthe supercritical fluid extraction and ascertain the best technological process by using the orthogonal test:Pressure 25MPa; Temperature 40℃; Timel20min; CO2 flux20L/g;The biggest extraction yield of fat: 57.03%.2. In the research it used the contrast experiment to determine the best cellbroken-down wall methodfor the ultrasonic wave broken, and the basis material determination ultrasonic wave broken craft conditionwas: Operating time is 5s;separated time is 5s;the supersonic number of times is 30;the temperature isnormal and the output is 400W.3. In. the research it takes withdrew in the corn embryo the return to original state GSH as the startingpoint, experiments four kinds of extraction methods successfully, namely the hot water extracted, theformic acid extracts, the ethyl alcohol extracts and the sulfuric acidextracts. The result indicated the hotwater extracts as the firstchoice method.4. The craft condition which raises to the hot water extraction carries on the comparison after theorthogonal experiment, determined the best hot water extraction raises the craft condition is: Solid fluidratiol:15g/ml; extraction temperature: 90℃; leaching liquor pH:3.0; extracted time 15min;the most greatlyextraction output of GSH in the hot water is the 91mg/100g of the corn embryo.5. It selects the PES membrane in this research to carryon ultra filters, and after the contrastexperiment, determines the best membrane aperture is 0.1μm, the best ultrafiltersthe pressure scope is:0.1-0.15MPa, protein temporarily stopping rate is 69.76%, the GSH returns-ratio is 90.35%.6. After the contrast experiment, it uses 732 cation resin to depurate the GSH solution, theconcentration of the eluant is the 0.2mol/L NaOH solution, the GSH maximum returns-ratio is 68.37%.7. The ultraviolet spectrophotometry and filter paper chromatography analysis is used in the researchto qualitate and quantitate the GSH, Compared with the standard preparation,make sure the extractedmaterial is GSH. According to the GSH content standarde quation of a curve to figure out the Content ofGSH is 1.25mg/100ml, namely in the corn embryo the extraction quantity of GSH is 56.21 mg/100g.8. The method of the isoelectric point is used to make GSH elutriant precip,the sediment is dissolvedafter centrifugalization and the content of GSH is 51.53mg/100g, the recovery is 91.67%. but the GSHextraction quantity of hot water is 91mg/100g, therefore in this experiment the extraction ratio of GSH is56.63%.9. In the research it uses the vacuum concentration to make the GSH concentrated and the watercontent is 40%, then use vacuum dehydration to obtain the cream color solid powder matter26.78mg, ofwhich the GSH is 25.77mg, namely the purity of GSH solid is 96.2%.
