Research Of The Extraction And Purification Methods Of Safflor Yellow

Safflor yellow has many physiological effects,and it has better stability compared with other natural pigments,it's application is increasingly broad,demand is growing too,so people requires Safflor yellow with high quality .The content of Safflor yellow in Safflower of Xinjiangis 22-30%,it is the good raw of Safflor yellow production.Against this we studied the separation of Safflor yellow, solved two problems in the production of Safflor yellow:low color value and high ash content.Isolated and identificated Safflor yellow,and established the HPLC fingerprint of Safflor yellow.Optimized the extraction process of Safflor yellow by orthogonal experiments : materials:liquid=1:12,extracts three times at 70℃and 1.5hr each time; Purified Safflor yellow by macroporous resin : materials: resin = 1:10, sample flow rate is 2.5 ml / min, eluting flow rate is 2.5 ml / min, the ratio of diameter and height of the resin column is 1:13; Isolated and identificated Safflor yellow by high preparative performance liquid chromatography, mass spectrometry and nuclear magnetic resonance,got two elements: Hydroxy Safformin A and Anhydrosafflor yellow B; The chromatography condition of HPLC fingerprint of Safflor yellow: the mobile phase: A-acetonitrile and B-0.5% phosphoric acid solution, gradient elution procedures: 0 ~ 60 min A (5% ~ 25%), B (95% ~ 75%), 60 ~ 70min A (25% ~ 5%), B (75% ~ 95%); detected wavelength :275 nm;velocity: 1 ml / min; temperature: 25°C; sample volume: 10μl.The color value of the Safflor yellow purified by the optimized process is beyond 1500 and ash content is down to 4%,the extractive yield is more than 11%.The tests verified that the process is rational and stable;The Safflor yellow purified by macroporous resin contains hydroxyl yellow A and many other active ingredients, it is good raw materials of foods, health products and medicine;The establishment of fingerprint of Safflor yellow provides a theoretical basis for the establishment of more scientific quality standards of Safflor yellow.
