The fruits of Gardenia has been used for the remedy for hepatic pain due to cirrhosis, abdominal pain due to dysentery, anti-phlogistics, diuretic and choleretic. Recent medicinal reports indicate that Gardenia herb also has sedative effect. The major active constituents of Gardenia fruits is geniposide. Gardenia pigments is also used as natural food color additive.In this paper, the macroporous adsorption resin and silical gel methods were applied to the separation and purification of geniposide and gardenia yellow from Gardenia successively, and the purity of the geneposide products obtained were 95.1%. The main experiment content is bellows:1 Determine the components of the Gardenia fruit. Establish a TLC-UV method to determine the geniposide from Gardenia fruit, and this method is economical and almost accurate. And it also is used as guidance when we research the extract experiment.2 Based on the extract content of geniposide and the gardenia yellow, do the monofactorial experiments and the orthogonal experiments. The best technologic extract of geniposide is: extraction solvent 60%(v/v) ethanol, temperature 20℃, duration 1 hour, and material/ethanol ratio 1∶25. The best technologic extract of gardenia yellow is: extraction solvent 60%(v/v) ethanol, temperature 40℃, duration 1 hour, and material/ethanol ratio 1∶20.3 Five kinds of macroporous resins were screened with regard to absorption capacity for geniposide and gardenia yellow. And the X-5 resin was determined to be the best one due to its relatively higher absorption and desorption capacity. The results also demonstrate that the effect of temperature is little to the absorption capability of X-5 resin. From dynamic absorption experiments of X-5, we can get the result below: 2-3 BV distilled water firstly can remove the most of carbonhydrates, amino and other polar substance; then 0.9BV 20%~35% ethanol can elute the geniposide while the gardenia yellow can't be eluted mostly; when the ethanol concentration surpass 40%, the gardenia yellow begins to be eluted.4 The geniposide product can be purified highly by the method of silical gel. The purity of geniposide can reach 92.3% with the yield of 80.3% when using the elute system of chloroform- methanol, while using gradient system of acetic ether–methanol, the purity and yield of the geniposide is 89.1%,79.3% respectively.5 The methods of TLC, HPLC-ESI-MS, and FT-IR were applied to determine geniposide product. All above approve that the product A4 is geniposide.
