Particle size of traditional Chinese medicine (TCM) is big, the size distribution is irregular, and quantity of active ingredient release is low and uneven. Stability and availability of TCM curative effect have been badly influenced by this problem. There are many advantages for TCM micronization, such as improving quantity of active ingredient release, saving the resource of medicinal materials, and so on. But from the point of view of quantity of active ingredient release and effective power consumption of comminuting, how the reasonable size range of different TCM is, should be farther researched. Therefore, in order to improve TCM curative effect, it is necessary and important to study the reasonable size range of different TCM.In the paper, Salviae miltiorrhizae Bge, a kind of rhizomatic TCM, was chosen as the research object. High performance liquid chromatography (HPLC) was used to detect the content of DanShenSu and Salvianolic acid B in water decoction solution. All-around considering of particle size of Salviae miltiorrhizae Bge, soaking time, decoction time and times for decoction, orthogonal test was designed to optimize the parameters in water extraction process. The result showed that the particle size is the most important factor for the content of DanShenSu and Salvianolic acid B. According to the influence degree, the order of the factors affecting the content from most to least is expressed as: times for decoction, decoction time and soaking time. The optimum water extraction process for Salviae miltiorrhizae Bge is: soaking for 60minutes and decocting 40minutes each time for 3 times.According to the optimum water extraction process, the content of DanShenSu and Salvianolic acid B with 10 size ranges were studied. The result showed that in the 10 particle size ranges, it is wrong that the smaller particle size, the higher content of Salvianolic acid B. Salviae miltiorrhizae Bge of 2.6~3mm in the optimum water extraction process, the content of Salvianolic acid B reach to the maximum, outside this size range, the contents were all reduced. In the study condition of this paper, the best particle size range of Salviae miltiorrhizae Bge was 2.6~3mm. In the paper, high performance capillary electrophoresis (HPCE) was used to analyze the water decoction solution of Salviae miltiorrhizae Bge. The HPCE separation was designed by consulting some references. Several reasons were listed to explain why the appearance peak of DanShenSu and Salvianolic acid B are not detected.
