Migold Asteraceae as a category of plants, are rich in lutein.The antioxidant function of lutein in the age-related macular degenerative diseases,arteriosclerosis,cancer and other diseases,the course of treatment,plays an important role.The lutein oxidation resistance function has been more and more by people valued.Therefore,lutein extraction plants have practical significance.The paper first uses the organic solvent extraction separation lutein in marigold and carries on the content determination,secondly inspected the temperature,the illumination and the air to lutein stable influence, finally has compared the lutein and Vc oxidation resistance activenessExperiment with n-hexane as the extraction solvent,and carries on backflow extraction,ultrasonic extraction and water extraction of lutein in marigold.obtained absorbtance value of extract from these three extraction method to be possible to know,ultrasonic extraction and water extraction is better than backflow extraction.Respectively will be extracting solution of ultrasonic extraction and water extraction carries on saponification to deal with,through HPLC after saponification of the sample to be analyzed.ODS C18 column using column packing (4.6mm×250mm, 5μm) , mobile phase: A (acetonitrile: methanol = 9:1) , C: ethyl acetate.Detection time: 22min,the samples using gradient elution, 0min~5min when mobile phase C for 10%,5min~15min when mobile phase C 15%,15min~20min when mobile phase C for the 0% .10μl sample.By the experimental results,we can see that water extracted from the yield of lutein higher than ultrasonic extraction yield.Proved use n-hexane solvent 16h immersion,as extraction lutein in marigold powder particles best way.Through the lutein standard chromatogram and sample chromatogram comparing to prove after saponification of the samples obtained for lutein.Inspected the temperature,the illumination and the air to the lutein stable influence.The temperature regarding the lutein influence was along with the temperature unceasing increase,lutein has had the degeneration and the discoloration.Therefore,lutein should be carried on the experiment and the preservation under the low temperature condition.The illumination regarding the lutein influence was along with the illumination unceasing growth,the lutein absorbance values reduces unceasingly , showed the lutein the chemical constitution changes.Therefore,lutein should be carried on the experiment and the preservation under dark condition. Lutein absorbance values in the air show, The air is opposite in the xanthophyl stable influence is bigger. Along with exposes in the air time extension, its absorption value drops obviously. Therefore, lutein must be preservation in cold, dark, obturation conditions.FRAP method used for antioxidant activity of lutein research. use microplate reader measured the samples absorbance values,calculate the lutein anti-oxidation value and Vc antioxidant activity compared to examine lutein in samples of antioxidant. The results showed that the sample concentration in range of 0.002 mg / ml~0.008 mg / ml , The lutein anti-oxidation stronger than the Vc anti-oxidation. therefore lutein is a better antioxidant effect with the drug, lutein has the good prospects for development.
