| Marigold is a common composite plant, rich in lutein. The function of antioxidant luteinplays an important role in the treatment of age-related macular degeneration, diabetes,cardiovascular disease, cancer, based on a large number of basic researches and clinicalpractice. In this thesis, we will research the extraction, quality standards, antioxidant activityevaluation of lutein in marigold pollen grains.1. Inspection of Lutein extraction process optimizationSmash dried marigold pollen particles, and soak with four times amount of n-hexaneovernight to get the crude extract. Charge nitrogen into the crude extract and do thesaponification for6hours, then extract with ethyl acetate, dehydrate, and concentrate intoextractum, then wash with hexane, get centrifugated for10min, precipitated, then turn out tobe lutein sample, with the yield rate as34%. Extract with ethyl acetate hexane for severaltimes, wash repeatedly to achieve the purpose of purification and removing impurity, toincrease the content of lutein. Determine the content of the purified product by HPLCchromatography to. The result shows that the content of lutein could be up to80%.This thesis investigates the influence the different extraction time, different extractiontemperature on the absorbance of lutein esters. The absorbance of lutein esters extractedunder different conditions of UV spectrophotometer, and draw the curve, to filter out the bestextraction time is about18h, the optimum extraction temperature is24℃. The soak theMarigold extraction time, soak temperature, saponification time, saponification temperaturefor the investigated factors, the orthogonal design to optimization of lutein extraction process,and the orthogonal experiment results for statistical analysis, screening out of lutein superiorextraction process for the12h of extraction time, extraction temperature24℃, saponificationtime3h, saponification temperature of90℃.2. The study on the standard of quality of LuteinAccording to the quality standard on traditional Chinese medicine of Ch.P2010, thisthesis establishes a standard of the quality of lutein and observes the precision, therepeatability, the stability, the linearity of lutein, the recoveries and the minimum detection limits by HPLC. This thesis provides a feasible standard on the control of drug quality, andfills the gap of the standard of the quality of lutein.3. The evaluation on the antioxidant activity of LuteinThis thesis calculates the antioxidant value of lutein samples and evaluates theantioxidant activity of lutein in contrast with that of Vc and BHT. The results show that theantioxidant activity of lutein is strong when the concentration is in the range of0.05mg/mL~0.50mg/mL. This experiment not only provides the basis of the study of thepharmacology of Lutein, but also lays a foundation of the development of Lutein drugs. |
