Visual fatigue is a kind of chronic syndrome based on eye symptoms, which has already became an interference and damage to the body and mind health of people. Now, drugs intervention is one of the common traditional treatment methods, while it exists large side effects, so the development of a kind of beverage, which has the effect of alleviating visual fatigue and based on the medicine food homology plant raw material, makes a big significance. Lutein is internationally recognized as "eye nutrients," its protective effect of retinal and alleviation effect of eye strain have been proved by many research. For our study, we isolated lutein from marigold and modified it into a water-soluble pigment. Then, we mixed it with a leaching solution of traditional plant materials that included Chinese wolfberry, chrysanthemum and cassia seed to develop a functional beverage and investigated its effects on visual fatigue alleviation in rats. The results of the study are as follows:Organic solvent extraction ultrasonic auxiliary is determined the best extraction method, the optimized extraction conditions was obtained by orthogonal experiment optimization and adjustment of the ultrasonic wave assisted, that is the liquid-solid ratio for rejoice is30:1, extraction time is40min, extraction temperature is40℃.On this condition, the lutein of Marigold is3.96mg/g, the concentration of purified lutein is50.12mg/100g.The optimal formula is obtained through orthogonal experiment:the content of Chinese wolfberry, chrysanthemum and cassia seed is a3,b1,0.08%, the content of sugar,citric acid and lutein is10%,0.10%,C2. Through constant temperature accelerated test,we predict the shelf life of this beverage is12months, the hygiene, sensory, physical, chemical and microbiological indexes of the beverage were within the scope of the national standard during this period.Lutein was stable to heat and alkaline, faintly acid solution, while it sensitive to light. Vc has certain protective effect on lutein, Fe3+, Fe2+, Cu2+has a strong damage to lutein, therefore, should try to iron and bronze should be avoided in the process of preparation and use of lutein beverage.To obtain rats of the syndrome of visual fatigue, high blood sugar model was built in the animal experiment, compared with model group, It is obvious to see that the lens injuries were alleviated in a dose-dependent manner by the lutein-rich beverage. Moreover, lutein-rich beverage can reduce free radical damage to the cells of the liver by protecting the SOD, CAT, GPx, and GR antioxidant enzyme systems, and enhance the regulatory ability of the eye muscles to aid recovery from visual fatigue.A comprehensive assessment of oxidation resistance effect of the beverage was conducted by combining in vivo and in vitro experiments. The scavenging activity of lutein beverage on hydroxyl radicals (·OH), oxygen anion (O2-·), DPPH· were studied, results indicated that lutein beverage had relatively high antioxidant activity. Moreover, lutein beverage can remarkably prolong the replication of K6001yeast.
