Seabuckthorn is a valuable plant to the treasure trove of medicinal plants in our country, its fruits not only contain a wide range of vitamins, microelements, amino acids, fatty acids and other biologically active substances, but also contain a variety of essential mineral elements to human, especially the most content is calcium,iron,zinc,potassium and selenium, it can be widely used in food, medicine, light industry, aerospace, agriculture, animal husbandry, fisheries and foreign trade in many sectors of the national economy. Our country has abundant resource of the Seabuckthorn, a large number of Seabuckthorn fruit used in the production of fruit juice or oil every year, but the marc that has abundant flavonoids was regarded as wastes. It was greatly a waste of resource. The Seabuckthorn marc from Qinghe in Xinjiang is raw material of this research. This paper has involved flavonoids extraction, separation and application in Seabuckthorn marc.On base of the previous studies, the flavonoids of Seabuckthorn were determinated by spectrophotometer and high-performance liquid chromatography in this paper. Using ethanol as solvent, it compared conventional solvent extraction with ultrasound-assisted method on the flavonoids of marc Seabuckthorn extraction. The optimum technological conditions of conventional solvent extraction were attained by single factor and orthogonal experiment as follow: ethanol concentration 60%, extraction temperature 70℃, the ratio of solid to liquid 1:40, extraction time 2h; the optimum conditions of ultrasound-assisted were: ethanol concentration 50%, ratio of solid to liquid 1:25, extraction time 20 min, the power of ultrasonic 0.4 kW.The results showed that: ultrasound-assisted extraction can be greatly reduced solvent consumption and extraction time compared two methods. It showed that the compositions of Seabuckthorn flavonoids included Catechin, Rutin, Quercetin and Isorhamnetin by determination of the high-performance liquid chromatography. Using polyamide column chromatography on the separation and purification of flavonoids, five components have been extracted from the ethyl acetate phase, compare with the retention time of standard by high-performance liquid chromatography, a preliminary determination as follow: componentⅠfor Rutin, componentⅣfor Quercetin, componentⅤforIsorhamnetin. Finally, the flavonoids as functional factor added to the milk for thedevelopment of functional milk tablets, it has the promotion of high value for thenutritional value of milk tablets and the health role of flavonoids.The extraction technology adopted in this paper is simple and the cost of manufactures islow, whereas the results showed the yield of the flavonoids was high. Producing milktablets of Seabuckthorn flavonoids used the process conditions in this article provides anew way for the processing and utilization of Seabuckthorn flavonoids and has a wideindustrialized foreground.
