With the aim of extraction, separation and purification of flavonoid from Fructus Aurantii Immaturus, the technological condition on extracting flavonoid through ultrasonic assist was studied in this paper, and the structures of flavonoids were confirmed with magnesium hydrochloric acid, UV, IR and so on.(1) Identification, determination of total flavonoids. In this paper, flavonoids were extracted from Fructus Aurantii Immaturus. Flavonoids were identified by reacting with magnesium hydrochloric acid, UV and IR characterization. All the flavonoids in Fructus Aurantii Immaturus were determined by spectrophotometry with the flavonoids which was synthesized by the teacher as the standard The method was simple and shortcut, and it provided a rapid and effective evaluation method to explore the best extraction and purification process.(2) Extraction of flavonoids from Fructus Aurantii Immaturus. Through a single-factor test, the mathematical model was established through Box-Beheken design, on the basis of the optimization of the processing time, power, temperature with ultrasonic-assist and the extraction rate of flavonoids. Variance analysis of the regression model indicated that:the regression equation was significant with good fitting degree. One degree term and second degree term have significant effort on the extraction rate of flavonoids, while the interaction between various factors has significant effort:the material to water has the greatest effort on the extraction rate of flavonoids under ultrasonic-assisted extraction condition, followed by the temperature, power and time. According to this model, the extraction rate of flavonoids 3.51% in the condition of ultrasonic-assisted time 15min, ultrasonic-assisted power 60%, ultrasonic-assisted temperature 62℃(3) Purification of total flavonoids from Fructus Aurantii Immaturus.①The activated carbons which were prepared from Fructus Aurantii Immaturus were under the conditions of heating temperature of 450℃, heating-up rate of 3℃/min, heat preservation temperature of 500℃and heat preservation time of 30 min, particle size of 0.45~4mm, had methylene blue specific surface area of 1477m2/g, hydroxyl was 2.3 m mol/g, carbonyl was 1.2 m mol/g total acid was 3.5 m mol/g.②According to the analysis of adsorption characteristics of activated carbon from the Fructus Aurantii Immaturus, it was found that it had the chracteristics of adsorption isotherm. The results showed that activated carbon from Fructus Aurantii Immaturus had higher absorption and desorption capacity. The absorption rate of Naringin, Hesperidin, Neohesperidin was 71.7%,57.4%,67.3%。The desorption rate of Naringin, Neohesperidin was 27.9%,78.4%.The optimum technological parameters of the purification of flavonoids by activated carbon from the Fructus Aurantii Immaturus are as follows:liquid ratio of 1:12, absorption time 3 h, absorption temperature 30℃, and methanol as the desorption solution, desorption time 3h, desorption temperature 30℃...
