In this paper,the measuring method of pectinase activity by an ultraviolet spectrophotometer was explored.The scouring process and the one-bath desizing-scouring process of cotton fabrics withα-amylase and pectinase were also studied.The research findings have great significance on the rapid test of the pectinase activity in water solution and promoting the cotton fabrics pretreatment process.Firstly,the feasibility and accuracy of the measuring method of pectinase activity with ultraviolet spectrophotometry method were explored.The characteristic absorption peak in ultra-violet region of pectin hydrolysate was measured.The characteristic absorptions of other chemical reagents involved in the process of hydrolysis of pectin by the pectinase were studied.The results showed that only the pectin hydrolysate has characteristic absorption in ultra-violet region.As compared with ultraviolet spectrophotometry method,the pectinase activity was measured with DNS method,It showed that the ultraviolet spectrophotometry method is much easier. more accurate and without any additive effect.In addition,there is no vigorous chemical reactiorl during the whole process.Therefore,this method could reflect the enzyme inactivation with little error and be a rapid measuring method of pectinase activity.Secondly.the hydrolysis effect of pectin in cotton fibers with two pectinases bv carbazole spectrophotometric method was studied.The results showed that ditTerent pectinases act on different substrates.Pectinase A could hydrolyze pectin from apples and had no hydrolytic action on the pectin in cotton fibers.It is just opposite for pectinase B.The pectin in cotton fibers as substrate,the activity of pectinase B could be measured with carbazole spectrophotometric method.The pectinase steam scouring process of cotton fabrics was optimized by t by 'change-one-factor-at-a-time' method.And the process was adjusted according to the practical need.Moreover,the effect of surfactant on the treatment was discussed.The results showed that the optimum process of pectinase steam process was as followed: solution(pectinase 2g/L.JFC 1g/L.pH=7)→2 dip-squeeze(100%)→steaming 30min at 90℃→boiling 20min in peregal O solution(2g/L)→water-washing.During pretreatment,surfactant could remove the wax on cotton fabric effectively and improve wettability of the cotton fabric greatly.Lastly,the effects of theα-amylase and pectinase on each other were analyzed when they were mixed as a composite.The one-bath desizing-scouring process of cotton fabrics withα-amylase and pectinase were optimized and adjusted.The final experiment process was as follows:solution(α-amylase 0.5g/L,pectinase 1g/L,JFC 1g/L)→2 dip-squeeze(100%)→steaming 20min at 100℃(100%humidity)→boiling 20min in peregal O solution(2g/L)→water-washing.
